木犀草素对动物卵巢氧化损伤的改善作用OA
The protective effects of luteolin against oxidative damage to the ovaries
[目的]探讨木犀草素对3-硝基丙酸诱导卵巢氧化应激损伤的改善效果,提升其在畜禽生产中增强繁殖性能的应用潜力.[方法]通过腹腔注射3-硝基丙酸(20 mg·kg-1)考察卵巢氧化应激模型的建立.在此基础上,将40只5周龄雌性小鼠随机分为对照组、模型组,以及木犀草素低剂量组(25 mg·kg-1)、中剂量组(50 mg·kg-1)和高剂量组(100 mg·kg-1).木犀草素预处理3周,并于第2~3周给予3-硝基丙酸造模.试验结束后测定雌性小鼠生长性能、卵巢指数、卵巢组织形态、抗氧化指标(总抗氧化能力、超氧化物歧化酶活性、过氧化氢酶活性、谷胱甘肽质量摩尔浓度,丙二醛质量摩尔浓度)及血清激素水平(抗缪勒管激素、雌二醇).[结果]与对照组相比,模型组卵巢指数极显著降低(p<0.01),成熟卵泡数量极显著减少(p<0.01),闭锁卵泡数量极显著增加(p<0.01),卵巢抗氧化能力显著下降(p<0.05),表明3-硝基丙酸成功诱导卵巢氧化应激损伤.在浓度筛选试验中,与模型组相比,木犀草素干预可不同程度改善卵巢氧化损伤;其中,中剂量组效果最为明显,表现为卵巢指数非常显著升高(p<0.001),成熟卵泡数量非常显著增加(p<0.001),闭锁卵泡非常显著减少(p<0.001),超氧化物歧化酶活性和总抗氧化能力极显著升高(p<0.01),同时丙二醛质量摩尔浓度极显著降低(p<0.01).各剂量木犀草素干预均可显著提高血清中抗缪勒管激素和雌二醇水平(p<0.05).[结论]木犀草素预处理能够有效缓解3-硝基丙酸诱导的卵巢氧化应激损伤,改善卵巢储备功能并促进卵泡发育,在缓解卵巢氧化损伤、维持卵巢功能方面具有潜在应用价值.
[Objective]This study investigated the ameliorative effects of luteolin(LUT)on 3-nitropro-pionic acid(3-NPA)-induced ovarian oxidative stress injury,aiming to enhance its application poten-tial for improving reproductive performance in livestock production.[Method]An ovarian oxidative stress model was established by intraperitoneal injection of 3-NPA(20 mg·kg-1).Based on this model,forty 5-week-old female C57BL/6J mice were randomly assigned to a control group(CK),a model group(MG),and three luteolin treatment groups receiving low(25 mg·kg-1),medium(50 mg·kg-1),and high(100 mg·kg-1)doses,respectively.Luteolin was administered as a pretreatment for 3 weeks,and 3-NPA was injected during weeks 2 to 3 to induce ovarian oxidative stress injury.At the end of the experiment,growth performance,ovarian index,ovarian histomorphology,antioxidant parameters(T-AOC,SOD,CAT,GSH,and MDA),and serum hormone levels(AMH and E2)were measured.[Result]Compared with the CK group,the MG group showed a highly significant decrease in ovarian index(p<0.01),reduced numbers of mature follicles(p<0.01),increased numbers of atretic follicles(p<0.01),and a significant decrease in ovarian antioxidant capacity(p<0.05),indicating that 3-NPA successfully induced ovarian oxidative stress injury.In the dose screening experiment,compared with the MG group,luteolin treatment alleviated ovarian oxidative damage to varying degrees,with the medium-dose group(50 mg·kg-1)showing the most pronounced effect.Specifically,the ovarian index was markedly increased(p<0.001),the number of mature follicles was increased and the number of atretic follicles was markedly reduced(p<0.001),SOD activity and T-AOC levels were significantly elevated(p<0.01),and MDA molaity was significantly decreased(p<0.01).In addition,all luteolin-treated groups showed significantly increased serum AMH and E2 levels(p<0.05).[Conclu-sion]Luteolin pretreatment effectively alleviated 3-NPA-induced ovarian oxidative stress injury,improved ovarian reserve function and promoted follicular development,demonstrating potential appli-cation value of luteolin in alleviating ovarian oxidative damage and maintaining ovarian function.
