白千层黄酮类化学成分及其抗菌活性OA

Flavonoid Constituents from Melaleuca leucadendron and Their Antibacterial Activities

本文对热带药用植物白千层(Melaleuca leucadendron Linn)中的黄酮类化学成分进行分离纯化鉴定并评价其抗菌活性.利用正相硅胶柱层析、Sephadex LH-20凝胶柱层析、半制备HPLC等色谱分离纯化该植物乙醇提取物中的黄酮类化学成分;并运用NMR以及MS技术鉴定化合物的结构.从白千层中分离鉴定了15个黄酮类化合物,分别为europetin 3-O-α-L-rhamnopyranoside(1)、杨梅苷(2)、槲皮素-3-O-鼠李糖苷(3)、杨梅素3-O-(3"-O-没食子酰)-a-吡喃鼠李糖苷7-甲基醚(4)、杨梅素3-O-(2"-O-没食子酰基)-a-吡喃鼠李糖苷7-甲基醚(5)、杨梅素3-O-(2"-O-没食子醇)-a-L-吡喃鼠李糖苷(6),槲皮素-3-O-β-D葡萄糖苷(7)、(-)-表没食子儿茶素3-O-没食子酸酯(8)、表儿茶素没食子酸酯(9)、(-)-没食子儿茶素3-O-没食子酸酯(10)、8-甲基-5,7-二羟基二氢黄酮(11)、乔松素(12)、槲皮素(13)、(-)-表没食子儿茶素(14)、没食子儿茶素(15).除化合物2和13外,其余化合物均为首次从白千层属植物中分离得到.此外,本文首次报道了化合物1的氢谱碳谱数据.抗菌活性测试表明化合物8~10对4种人体致病菌具有显著的抑制活性.

This study focused on the isolation,purification,identification,and antimicrobial activity evaluation of flavonoids from the tropical medicinal plant Melaleuca leucadendron Linn.The flavonoid constituents in the ethanol extract of this plant were separated and purified using various chromatographic techniques,including normal-phase silica gel column chromatography Sephadex LH-20 gel column chromatography,and semi-preparative HPLC.The structures of the isolated compounds were identified by NMR and MS techniques.A total of fifteen flavonoids were isolated and identified from M.leucadendron:europetin 3-O-α-L-rhamnopyranoside(1),myricitrin(2),quercetin-3-O-rhamnopyranoside(3),myricetin 3-O-(3′′-O-galloyl)-α-rhamnopyranoside 7-methyl ether(4),myricetin 3-O-(2′′-O-galloyl)-α-rhamnopyranoside 7-methyl ether(5),myricetin 3-O-(2′′-O-galloyl)-a-L-rhamnopyranoside(6),quercetin-3-O-β-D-glucoside(7),(-)-epigallocatechin 3-O-gallate(8),epicatechin gallate(9),(-)-gallocatechin 3-O-gallate(10),8-methyl-5,7-dihydroxydihydroflavonoid(11),pinocembrin(12),quercetin(13),(-)-epigallocatechin(14)and gallocatechin(15).Except for compounds 2 and 13,all other compounds were isolated from the genus Melaleuca for the first time.Additionally,the 1H and 13C NMR data of compound 1 are reported here for the first time.Antimicrobial activity tests showed that compounds 8-10 exhibited significant inhibitory activity against four species of pathogenic bacteria.
