Study on Terpenoid Constituents from Tanacetum tatsienense
本文采用系统溶剂提取法、各种色谱技术(正反相硅胶柱层析以及半制备高效液相色谱等)对打箭菊(Tanacetum tatsienense)乙酸乙酯提取物的化学成分进行分离纯化.通过各种波谱技术(1D和2D-NMR)、高分辨质谱、量子化学的计算以及化学方法对所分离的化合物进行结构鉴定,分离并鉴定6个萜类化合物,其中化合物1为新去胡萝卜素苷,命名为tatsienenoside A(1),其余的已知化合物分别鉴定为3β-hydroxy-β-ionone(2)、1β-hydroxy-7-epi-costic acid methyl ester(3)、阿里二醇(4)、款冬二醇(5)、马尼拉二醇(6).采用CCK-8方法对所分离的萜类化合物进行体外抗肿瘤细胞增殖活性测试,结果表明阿里二醇对人非小细胞肺癌A549细胞有较好的抗增殖活性,IC50值为(12.58±0.60)µmol/L.
In this study,systematic solvent extraction techniques and different chromatographic methods,such as normal-phase and reverse-phase silica gel column chromatography,along with semi-preparative high-performance liquid chromatography,were employed to isolate and purify the chemical components extracted from the ethyl acetate extract of Tanacetum tatsienense.The structures of the obtained compounds were determined using a range of spectroscopic techniques,including 1D and 2D NMR,high-resolution mass spectrometry,quantum chemical calculations,and conventional chemical methods.Six terpenoids were isolated and identified.Among them,compound 1 was recognized as a new norcarotenoid glycoside,named as tatsienenoside A(1).The remaining compounds were identified as known terpenoids,including 3β-hydroxy-β-ionone(2),1β-hydroxy-7-epi-costic acid methyl ester(3),arnidiol(4),faradiol(5),and maniladiol(6).The anti-proliferative activity of the isolated terpenoids was assessed using the CCK-8 method.The results indicated that arnidiol(4)exhibited moderate anti-proliferative activity against human non-small cell lung cancer A549 cells,with an IC50 value of(12.58±0.60)μmol/L.
