目的:通过急性毒性、最大耐受量及亚急性毒性试验,评价蒲芩解毒利湿颗粒的毒理学安全性,为其临床安全应用提供理论依据.方法:选取40只雌雄各半的健康昆明(KM)小鼠,分三组开展急性毒性试验,单次灌胃给药后连续观察7天,记录体重、精神状态及死亡情况;选取20只雌雄各半的 KM 小鼠进行最大耐受量试验,以 0.52 g/mL 浓度药液、40 mL/kg 体积灌胃,连续观察 14 天,记录中毒与死亡情况;选取 80 只雌雄各半的健康 SD 大鼠,随机分为对照组及高(6.4 g/kg)、中(3.2 g/kg)、低(1.6 g/kg)剂量组,灌胃给药后观察大鼠临床状态,检测血常规、血清生化指标,计算脏器系数并做HE染色病理检查.结果:急性毒性和最大耐受量试验中,各组小鼠7天内无死亡,精神、体重均正常,最大耐受量大于20.8 g/kg,组间无统计学差异(P>0.05);亚急性毒性试验中,各组大鼠4周内无异常死亡,血常规、生化指标及脏器系数均正常(P>0.05),脏器病理检查无明显异常改变.结论:蒲芩解毒利湿颗粒安全性良好,无明显急性及亚急性毒性,可进一步开发为慢性尿路感染治疗药物,为相关患者提供新的治疗选择.

Objective:This study aimed to evaluate the toxicological safety of Puqin Jiedu Lishi granules through acute and subacute toxicity tests,and to provide a theoretical basis for its safe clinical application.Methods:Forty healthy KM mice(half male and half female)were divided into three groups for the acute toxicity test.All mice received a single intragastric administration,followed by a 7-day observation of body weight,mental state and mortality.Twenty KM mice(half male and half female)were subjected to the maximum tolerated dose test.The drug solution(0.52 g/mL)was administered at 40 mL/kg,with a 14-day observation of toxic reactions and mortality.Eighty healthy SD rats(half male and half female)were randomly assigned to the control group,as well as high-dose(6.4 g/kg),medium-dose(3.2 g/kg),and low-dose(1.6 g/kg)groups.After intragastric administration,the general clinical status of rats was monitored,blood routine parameters and serum biochemical indices were determined,organ coefficients were calculated,and histopathological alterations were examined via HE staining.Results:No death or abnormal manifestations were found in mice during acute toxicity and maximum tolerated dose tests,with a maximum tolerated dose over 20.8 g/kg and no significant inter-group differences(P>0.05).In the subacute toxicity test,all rats survived normally within 4 weeks;blood routine,biochemical indexes and organ coefficients were normal(P>0.05),and no obvious histopathological lesions were observed.Conclusions:Puqin Jiedu Lishi granules show favorable safety with no obvious acute or subacute toxicity,and are worthy of further development as a therapeutic agent for chronic urinary tract infections,providing a new treatment option for patients with such diseases.
