人参皂苷Rg1对DSS诱导溃疡性结肠炎小鼠肠道屏障及菌群的调控作用OA
Regulatory effect of ginsenoside Rg1 on intestinal barrier and gut microbiota in mice with DSS-induced ulcerative colitis
目的:探讨人参皂苷Rg1 对葡聚糖硫酸钠(DSS)诱导的溃疡性结肠炎(UC)小鼠肠道屏障功能与肠道菌群稳态的调节作用及机制.方法:采用雄性SPF级C57BL/6小鼠建立DSS诱导的UC模型,随机分为空白对照组、模型对照组及Rg1低[50 mg/(kg·d)]、中[100 mg/(kg·d)]、高[200 mg/(kg·d)]剂量组,连续灌胃给药14天.监测小鼠体重、粪便性状及便血情况并计算疾病活动指数(DAI);测量结肠长度,HE 染色评估结肠病理损伤;ELISA 检测结肠组织炎症因子水平;16S rDNA 高通量测序分析结肠内容物肠道菌群.结果:Rg1 可显著减轻 DSS 诱导的小鼠体重下降、降低 DAI 评分、恢复结肠长度(P<0.01),显著下调结肠组织 IL-1β、IL-6、TNF-α 及MPO 水平(P<0.01);减轻结肠黏膜损伤与炎症浸润,增加杯状细胞数量;提升肠道菌群 α 多样性,恢复菌群结构,纠正门、属水平菌群丰度异常,上调有益菌丰度.结论:Rg1 可有效缓解DSS诱导的小鼠UC症状及炎症反应,其机制与修复受损肠道屏障、调节失衡肠道菌群密切相关,为Rg1 治疗UC提供了实验依据.
Objective:To investigate the regulatory effects and underlying mechanisms of Rg1on intestinal barrier function and gut mi-crobiota homeostasis in mice with DSS-induced UC.Methods:Male SPF-grade C57BL/6 mice were used to establish the DSS-induced UC model,and were randomly divided into the blank control group,model control group,and Rg1 low-dose[50 mg/(kg·d)],medium-dose[100 mg/(kg·d)]and high-dose[200 mg/(kg·d)]groups,with intragastric administration once daily for 14 consecutive days.The body weight,fecal traits and hematochezia of the mice were monitored,and the DAI was calculated.Colon length was measured,colonic pathological damage was evaluated by HE staining,the levels of inflammatory factors in colon tissues were detected by ELISA,and the intestinal microbiota in colonic contents were analyzed via 16S rDNA high-throughput sequencing.Results:Rg1 significantly alleviated DSS-induced body weight loss,re-duced DAI score,and increased colon length in mice(P<0.01).It also markedly down-regulated the levels of IL-1β,IL-6,TNF-α and MPO in colon tissues(P<0.01),attenuated colonic mucosal injury and inflammatory infiltration,increased goblet cell count,elevated intestinal micro-biota α-diversity,restored microbial community structure,corrected abnormal flora abundance at phylum and genus levels,and up-regulated the abundance of beneficial bacteria.Conclusions:Rg1 can effectively alleviate the symptoms and inflammatory response of DSS-induced UC in mice,and its mechanism is closely related to repairing the impaired intestinal barrier and regulating the dysregulated intestinal microbiota.This study provides experimental evidence for the clinical application of Rg1 in UC treatment.
