目的 以网络药理学联合分子对接技术,系统阐释芦根-薏苡仁对药防治胸外科术后肺部感染的作用机制.方法 经TCMSP、PubChem等多数据库筛选获取靶点韦恩图,构建中药-成分-靶点网络图,进行GO和KEGG富集分析,并进行分子对接验证.结果 经筛选获得10个活性成分(如Stigmasterol、2-Monoolein等),预测靶点252个;整合GeneCards等数据库获得疾病靶点2 627个,取交集得127个"药物-疾病"关键靶点.STRING构建蛋白互作网络并拓扑分析,锁定MAPK3、JAK1、PRKCD等10个核心靶点.GO及KEGG富集显示,其作用主要涉及G蛋白偶联受体信号、钙离子调控、磷脂酶D及鞘脂信号通路等.分子对接结果表明,核心成分与靶点结合能均<-5 kJ/mol,结合稳定.结论 芦根-薏苡仁通过"多成分-多靶点-多通路"协同发挥抗炎及免疫调节作用,为该对药的现代化研究与临床转化提供理论依据.

Objective To systematically elucidate the mechanism of Rhizoma Phragmitis-Semen Coicis couplet medicines in preventing and treating postoperative pulmonary infection in thoracic surgery using Network Pharmacology combined with molecular docking technology.Methods Targets were screened using multiple databases including TCMSP and PubChem to obtain Venn diagrams.A Chinese medicine-component-target network was constructed,followed by GO and KEGG enrichment analysis and molecular docking verification.Results A total of 10 active components(e.g.,Stigmasterol,2-Monoolein,etc.)were screened,with 252 predicted targets,2 627 disease targets were obtained by integrating databases such as GeneCards,and 127 key drug-disease intersection targets were identified.A protein-protein interaction(PPI)network was constructed by STRING and subjected to topological analysis,identifying 10 core targets including MAPK3,JAK1,and PRKCD.GO and KEGG enrichment analyses revealed that the effects were mainly involved in G protein-coupled receptor signaling,calcium ion regulation,phospholipase D and sphingolipid signaling pathways,etc.Molecular docking results showed that the binding energies of core components to targets were all less than-5 kJ/mol,indicating stable binding.Conclusion Rhizoma Phragmitis-Semen Coicis couplet medicines exerts synergistic anti-inflammatory and immunomodulatory effects through a"multi-component,multi-target,multi-pathway"mode,providing a theoretical basis for the modern research and clinical transformation of this couplet medicines.
