微流控技术在居家核酸检测中的应用进展OA

核酸检测的应用场景多为医院,通常需要专业人员采样,并使用专业设备完成检测.随着微流控技术集成化程度提高以及核酸等温扩增技术的发展,居家场景下的核酸检测技术近年来发展迅速.受检者可在家庭场景中通过采集血液、唾液、尿液和粪便等样本,利用集成化微流控核酸检测系统开展自我健康检测,检测范围已涵盖常见的呼吸道病毒、人类免疫缺陷病毒(HIV)、人乳头瘤病毒(HPV)、肝炎和消化道病毒等.相比医院场景下开展的核酸检测,居家核酸检测具有简单方便、快捷和成本低廉等优点.此外,基于微流控技术的居家核酸检测方法还适合在缺乏基本医疗条件的地区开展核酸检测.本文对目前微流控技术在居家核酸检测中所使用的流体操控与扩增技术体系、不同样本类型的处理方法及应用情况进行了概述,总结了居家核酸检测对微流控技术提出的新要求和现存的技术挑战,并展望了基于微流控技术的居家核酸检测技术的未来发展方向.

Currently,nucleic acid testing is primarily used in hospitals,typically requiring specialized personnel using specialized equipment.With the increasing integration of microfluidics,the development of isothermal nucleic acid amplification,and a growing demand for a healthier lifestyle,nucleic acid testing at home has attracted increasing attention in recent years.Patients can now use microfluidic chips to conduct self-health tests at home,using the samples such as blood,saliva,urine,and stool.Current in-house nucleic acid testing covers common respiratory viruses human immunodeficiency virus(HIV),human papilloma virus(HPV),hepatitis,and digestive tract viruses.At the same time,in-house nucleic acid testing based on microfluidics is also suitable for remote areas lacking basic medical facilities.This study reviewed the current microfluidics techniques for nuclei acid home testing,sample types,amplification techniques,and examples of microfluidics in-house nucleic acid testing.This review also summarized the requirements and technical barriers for microfluidics in in-house nucleic acid testing and prospected the future development direction of in-house nucleic acid testing based on microfluidic techniques.
