耐低温降解木质纤维素芽孢杆菌的筛选及性质研究OA
Screening and characterization of lignocellulose-degrading Bacillus strains with low-temperature tolerance
[目的]筛选耐低温降解木质纤维素的芽孢杆菌菌株,以解决低温环境下木质纤维素类生物质资源化利用效率低下的问题.[方法]采集河北省献县秋江合作社白星花金龟养殖基地新鲜的白星花金龟粪便,进行菌株分离、初筛、复筛,对获得的菌株通过形态学观察、16S rDNA序列分析和生理生化验证试验进行种属鉴定,并在不同温度和pH条件下对其活菌数和纤维素酶活进行测定.[结果]从白星花金龟的粪便中筛选得到 5 株木质纤维素降解菌,分别为特基拉芽孢杆菌(Bacillus tequilensis)N5-2、解淀粉芽孢杆菌(Bacillus amyloliquefaciens)Y5-3、枯草芽孢杆菌(Bacillus subtilis)R4-3、暹罗芽孢杆菌(Bacillus siamensis)N3-5和死谷芽孢杆菌(Bacillus vallismortis)R4-4.R4-3 和R4-4 随着温度降低纤维素酶活力下降缓慢,R4-3 在 20℃时酶活力仍然在 80 U/mL以上,R4-3 在pH 4.0 时纤维素酶活达 80 U/mL.[结论]分离获得 5 株耐低温产木质纤维素酶的芽孢杆菌,其中枯草芽孢杆菌R4-3 菌株可在 10℃、pH 3.5 条件下生长,在 20℃、pH 4.0 条件下生长良好,且纤维素酶活力达 80 U/mL左右,为北方冬季农林废弃物的高效利用提供菌株资源.
[Objective]In order to address the issue of low resource utilization efficiency of lignocellulose-like biomass under low-tem-perature conditions,this study was conducted to screen Bacillus strains that can degrade lignocellulose at low temperatures.[Methods]The fresh fecal samples of Protaetia brevitarsis were collected from the breeding base of Protaetia brevitarsis at Xianxian Qiujiang co-operative in Hebei Province.Following the isolation,primary screening,and secondary screening of strains,the obtained isolates were subjected to species identification via morphological analysis,16S rDNA sequence analysis,and physiological and biochemical charac-terization assays.Additionally,their cellulase activities were determined under different temperature and pH conditions.[Results]Five lignocellulose-degrading strains were isolated and screened from the feces of Protaetia brevitarsis,named Bacillus tequilensis N5-2,Bacillus amyloliquefaciens Y5-3,Bacillus subtilis R4-3,Bacillus siamensis N3-5,and Bacillus vallismortis R4-4.The cellulase ac-tivities of R4-3 and R4-4 exhibited a slow decline with decreasing temperature.Specifically,the cellulase activity of R4-3 remained above 80 U/mL even at 20 ℃,and could also reach 80 U/mL at pH 4.0.[Conclusion]Five low-temperature-resistant Bacillus strains that produce lignocellulase were isolated.Among them,Bacillus subtilis R4-3 strain could grow at 10℃and pH 3.5,and grew well at 20℃and pH 4.0,with cellulase activity reaching about 80 U/mL,which provided strain resources for the efficient utilization of agri-cultural and forestry waste in northern winter.
