α-烯醇化酶与鼻黏膜巨噬细胞胞葬缺陷的相关性及其对嗜酸性粒细胞型慢性鼻窦炎伴鼻息肉的鉴别价值OA
ENO1 is associated with impaired nasal mucosal macrophage efferocytosis and discriminates eosinophilic chronic rhinosinusitis with nasal polyps
目的 明确α-烯醇化酶(α-enolase,ENO1)在慢性鼻窦炎伴鼻息肉(CRSwNP)中的表达与胞葬作用和炎症的关联,及其对嗜酸性粒细胞型CRSwNP(eosinophilic CRSwNP,ECRSwNP)诊断效能.方法 基于公共单细胞转录组数据集分析ENO1表达并采用实时荧光定量聚合酶链式反应(qRT-PCR)验证.提取胞葬相关巨噬细胞再聚类,比较ENO1高、低表达亚群的胞葬相关信号表达差异.Spearman相关性分析其与炎症细胞相关性;受试者工作特征(ROC)曲线评估其对ECRSwNP诊断效能.结果 ENO1在ECRSwNP中增加最为明显,且主要高表达于巨噬细胞.与ENO1低表达巨噬细胞相比,高表达细胞中胞葬信号(CX3CR1、MERTK、GAS6)明显降低.ENO1与组织嗜酸性粒细胞计数呈正相关趋势(r=0.32,P=0.051).ENO1预测ECRSwNP曲线下面积为0.706(95%CI:0.542-0.870,P=0.028),敏感度为71.43%,特异度为66.67%.结论 ENO1高表达与鼻黏膜巨噬细胞胞葬功能缺陷及炎症相关,且对ECRSwNP的鉴别具有中等诊断价值.
OBJECTIVE To determine the expression of α-enolase(ENO1)in chronic rhinosinusitis with nasal polyps(CRSwNP)and examine its relationships with macrophage efferocytosis and inflammation,and evaluate its diagnostic efficacy for distinguishing eosinophilic CRSwNP(ECRSwNP).METHODS We analyzed a public single-cell RNA-sequencing dataset to determine the expression level of ENO1 and predominant expressing cell types in CRSwNP,and validated ENO1 expression by qRT-PCR.Macrophage subsets related to efferocytosis were extracted and reclustered;differential expression of efferocytosis-related genes between ENO1-high and ENO1-low macrophages was assessed.Spearman correlation was used to examine associations between ENO1 expression and tissue inflammatory cell counts.Receiver operating characteristic(ROC)curves evaluated the ability of ENO1 to distinguish ECRSwNP.RESULTS ENO1 was most significantly elavated in ECRSwNP and prominently expressed in macrophages.Compared with ENO1 low-expressing macrophages,ENO1 high-expressing macrophages exhibited reduced expression of efferocytosis-related genes(CX3CR1,MERTK,GAS6).ENO1 showed a positive trend with tissue eosinophil counts(r=0.32,P=0.051).ROC analysis showed an area under the curve of 0.706(95%CI:0.542-0.870,P=0.028)for identifying ECRSwNP,with a sensitivity of 71.43%and a specificity of 66.67%.CONCLUSION ENO1 is upregulated in ECRSwNP,associates with impaired efferocytosis and inflammation,and shows moderate diagnostic value for distinguishing ECRSwNP.
