具有可注射性及抗菌性能的动态交联水凝胶OA
A Dynamically Crosslinked Hydrogel with Injectable and Antibacterial Properties
慢性伤口的临床管理常常受到细菌感染和生物膜形成的挑战.为解决这一问题,我们通过氨基化壳聚糖(amide-modified chitosan,AMCS)与氧化葡聚糖(oxidized dextran,ODex)之间的动态席夫碱交联,开发了一种可注射的水凝胶(AMCS@ODex).该交联策略不仅改善了壳聚糖固有的局限性,如溶解性和力学性能较差,还赋予了水凝胶优异的剪切稀化行为.AMCS@ODex 水凝胶对大肠杆菌和耐甲氧西林金黄色葡萄球菌(methicillin-resistant Staphylococcus aureus,MRSA)均表现出抗菌活性,抗菌率分别为58%和73%.此外,它还能有效破坏已形成的 MRSA 生物膜,破坏率约为50%.至关重要的是,该水凝胶展现出优异的生物相容性,使 L929 成纤维细胞的存活率大于 90%.AMCS@ODex 水凝胶集良好的抗菌性能、理想的力学性能及高生物相容性为一体,在对抗细菌感染等生物医学应用中前景广阔.
The clinical management of chronic wounds is frequently challenged by bacterial infection and biofilm formation.To address this,we developed an injectable hydrogel,denoted as AMCS@ODex,via dynamic Schiff base bonding between amide-modified chitosan(AMCS)and oxidized dextran(ODex).This crosslinking strategy not only mitigated the inherent limitations of chitosan,such as poor solubility and mechanical properties,but also endowed the hydrogel with excellent shear-thinning behaviors.AMCS@ODex exhibited antibacterial efficacy against Escherichia coli(E.coli)and methicillin-resistant Staphylococcus aureus(MRSA),and the antibacterial rates against E.coli and MRSA were 58%and 73%,respectively.Furthermore,it effectively disrupted pre-formed MRSA biofilms,leading to a disruption rate approximately 50%.Critically,the hydrogel exhibited outstanding biocompatibility,supporting over 90%viability of L929 fibroblast cells.By integrating good antibacterial performance,with desirable mechanical properties and high biocompatibility,the AMCS@ODex hydrogel presents potential biomedical applications,particularly in the fight against bacterial infections.
