为研究苏云金芽孢杆菌(Bacillus thuringiensis,Bt)对冷杉梢斑螟幼虫的致病力,在冷杉梢斑螟暴发的樟子松人工林内采集土样,经温度筛选法分离苏云金芽孢杆菌菌株后,通过0.5%碱性复红溶液(0.5g 碱性复红溶解于 20 mL 乙醇中,蒸馏水定容到 100 mL)染色进行形态鉴定,并采用 PCR-RFLP 鉴定体系和 vip3A 的特异性引物扩增方法对菌株所含 vip3A 和 cry 基因类型进行鉴定分析,最终结合 16SrRNA 测序和构建系统发育树确定了 5株 Bt 菌株,并测定各菌株对冷杉梢斑螟幼虫的致病性.结果表明:从土壤中分离得到 5 株 Bt 菌株,其中 BtM0003菌株含有 cry1Aa、cry9Ea 基 因;BtM0019、BtM0014 和 BtM0015 菌株均含有 cry1Aa、cry2Ab、cry9Ea、vip3Aa 基因;BtM0004 菌株含有 cry2Ab、cry9Ea、vip3Aa 基因.而冷杉梢斑螟幼虫在 5 株菌株处理下的累计校正死亡率由大到小依次为 BtM0015、BtM0014、BtM0019、BtM0004、BtM0003,其中 BtM0015 的累计校正死亡率最高为(88.33±7.64)%,半致死时间(LT50)为 3.866 d.

To investigate the pathogenicity of Bacillus thuringiensis(Bt)against larvae of Dioryctria abietella,soil samples were collected from a Pinus sylvestris var.mongolica plantation where D.abietella outbreaks occurred.Bt strains were isolated using the temperature screening method,followed by morphological identification via staining with 0.5%alkaline fuchsin solution.The types of vip3A and cry genes harbored by the strains were identified and analyzed using a PCR-RFLP identi-fication system and specific primer amplification for vip3A.Finally,five Bt strains were confirmed by 16SrRNA gene se-quencing and phylogenetic tree construction,and their pathogenicity to D.abietella larvae was determined.The results showed that five Bt strains were isolated from the soil.Among them,strain BtM0003 contained cry1Aa and cry9Ea,strains BtM0019,BtM0014 and BtM0015 all contained cry1Aa,cry2Ab,cry9Ea and vip3Aa,and strain BtM0004 contained cry2Ab,cry9Ea and vip3Aa.The cumulative corrected mortality of D.abietella larvae treated with the five strains ranked in descending order as follows:BtM0015>BtM0014>BtM0019>BtM0004>BtM0003.Among them,BtM0015 exhibited the highest cumulative corrected mortality of(88.33±7.64)%,with a median lethal time(LT50)of 3.866 days.
