参附注射液抑制NLRP3炎症小体介导的小胶质细胞焦亡减轻老年大鼠术后认知功能障碍OA
Shenfu injection inhibits NLRP3 inflammasome-mediated microglial pyroptosis and alleviates postoperative cognitive dysfunction in aged rats
目的 探讨参附注射液(SFI)对老年大鼠术后认知功能障碍(POCD)的影响.方法 采用异氟烷吸入麻醉联合开腹脾切除术建立老年POCD大鼠模型,随机分为假手术组、模型组、治疗组,每组24只.体外实验将培养的小胶质细胞分为空白组、脂多糖(LPS)组、LPS+SFI组.采用 Morris水迷宫(MWM)对大鼠进行行为 学 检 测,Western blot检测大鼠海马区组织核因子-κB(NF-κB)、NOD样受体热蛋白结构域相关蛋白3(NLRP3)、半胱氨酸天冬氨酸蛋白酶-1(Caspase-1)、凋亡相关斑点样蛋白(ASC)、GSDMD表达,ELISA检测大鼠血清IL-1、IL-6、高迁移率族蛋白B1(HMGB1)和肿瘤坏死因子-α(TNF-α)水平,HE染色、免疫荧光染色观察大鼠海马区组织炎症细胞浸润程度、荧光强度,扫描电子显微镜观察体外实验中小胶质细胞焦亡形态学变化.结果 与模型组比较,治疗组术后第 3、7天逃避潜伏期缩短(P<0.05),穿越原平台位置次数明显增多(P<0.05),术后第3天GSDMD、NF-κB、NLRP3、ASC、Caspase-1表达降低(P<0.05),术后第1、3、7天 HMGB1、IL-1、IL-6、TNF-α水平降低(P<0.05),炎症细胞浸润明显减轻,NF-κB、GSDMD表达降低.与LPS组比较,LPS+SFI组小胶质细胞的焦亡减轻.结论 SFI可能通过抑制NLRP3炎症小体介导的小胶质细胞焦亡减轻老年大鼠POCD.
Objective To investigate the effect of Shenfu Injection(SFI)on postoperative cognitive dysfunction(POCD)in aged rats.Methods An elderly rat model of postoperative cognitive dysfunction(POCD)was established by using isoflurane inhalation anesthesia combined with splenectomy.The rats were randomly divided into the sham operation group,the model group,and the treatment group,with 24 rats in each group.In the in vitro experiment,the cultured microglial cells were divided into the blank group,the li-popolysaccharide(LPS)group,and the LPS+SFI group.Behavioral assessment was performed using the Morris water maze(MWM),the expression of nuclear factor-κB(NF-κB),NOD-like receptor thermal protein domain associated protein 3(NLRP3),cysteinyl aspartate specific protease-1(Caspase-1),apoptotic speck-like protein(ASC),and GSDMD in the hippocampal tissue of the rats was detected by Western blot,the levels of IL-1,IL-6,high mobility group box 1(HMGB1),and tumor necrosis factor-α(TNF-α)in the rat serum were detected by ELISA,the degree and fluorescence intensity of inflammatory cell infiltration in the rat hippocam-pal tissue were observed by HE staining and immunofluorescence staining,and the morphological changes of pyroptosis of the microglial cells in the in vitro experiment were observed by scanning electron microscopy.Results Compared with the model group,the escape latency in the treatment group was shortened on the 3rd and 7th days after surgery(P<0.05)and significantly increased platform crossings(P<0.05).On the 3rd day after surgery,the expressions of GSDMD,NF-κB,NLRP3,ASC,and Caspase-1 decreased(P<0.05).On the 1st,3rd,and 7th days after surgery,the levels of HMGB1,IL-1,IL-6,and TNF-α decreased(P<0.05).The infiltration of inflammatory cells was significantly reduced,and the expression levels of NF-κB and GSDMD decreased.Compared with the LPS group,the pyroptosis of microglia in the LPS+SFI group was alleviated.Conclusion SFI may alleviate POCD by inhibiting the pyroptosis of microglia mediated by the NLRP3 inflammasome.
