目的 使用两样本孟德尔随机化方法探究干果摄入和2型糖尿病发病风险之间的因果关联.方法 基于全基因组关联研究汇总数据,以干果摄入为暴露因素,2型糖尿病为结局因素,进行两样本孟德尔随机化研究.主要分析方法为逆方差加权法(inverse variance weighting,IVW),分别采用F值和Cochran's Q统计量检验工具变量的稳定性和异质性.并采用MR-Egger回归、加权中位数法、基于众数的简单估计法、基于众数的加权估计法和留一法进行敏感性分析.结果 共纳入41个SNPs位点作为干果摄入的工具变量.孟德尔随机化的结果表明,干果摄入与2型糖尿病之间存在负向因果关系(IVW法OR:0.48,95%CI:0.30~0.76,P=0.002),且结果无多效性.加权中位数法和留一法的结果与IVW保持一致.结论 干果摄入与2型糖尿病发病之间存在负向因果关系.

Objective To explore the causal association between nut intake and the risk of type 2 diabetes mellitus with two-sample Mendelian randomization(MR).Methods Genome-wide association study(GWAS)-based pooled data were used to perform two-sample Mendelian randomization with nut intake as the exposure and type 2 diabetes mellitus as the outcome.The primary analysis method was inverse variance weighting(IVW).F-value statistics and Cochran's Q statistics were used to test the stability and heterogeneity of instrumental variables.MR-Egger regression,weighted median method,simple plurality-based estimation,weighted plurality-based estimation,and leave-one-out method were used for sensitivity analyses.Results A total of 41 single-nucleotide polymorphism loci were included as instrumental variables for nut intake.The results of Mendelian randomi-zation indicated an inverse causal association between nut intake and type 2 diabetes mellitus(IVW method OR:0.48,95%confidence interval:0.30-0.76,P=0.002),and there was no pleiotropy.The results obtained from the weighted median and leave-one-out methods were consistent with IVW.Conclusion There is a negative causal relationship between nut intake and the onset of type 2 diabetes mellitus.
