沉默RND1对舌鳞状细胞癌增殖、凋亡、侵袭和迁移的影响OA
Effects of RND1 knockdown on proliferation,apoptosis,invasion,and migration in tongue squamous cell carcinoma
目的 通过沉默RND1表达来评估其对舌鳞状细胞癌(tongue squamous cell carcinoma,TSCC)的细胞增殖、凋亡、侵袭和迁移能力的影响,探究RND1在TSCC发生发展中的功能作用.方法 采用Western blot和免疫组化检测TSCC组织及配对癌旁组织中RND1蛋白表达水平.通过siRNA介导沉默TSCC细胞系CAL-27细胞和SCC-25细胞,分为RND1沉默组、阴性对照组和空白对照组.采用CCK-8法检测细胞增殖,采用流式细胞术检测细胞凋亡情况,采用Tr-answell 实验和划痕实验检测细胞侵袭和迁移能力.采用Western blot分析上皮-间质转化(epithelial-mesenchymal transi-tion,EMT)相关蛋白及下游信号通路相关蛋白的表达变化.结果 RND1蛋白在TSCC组织中的表达水平明显高于癌旁正常组织,P<0.05.与空白对照组和阴性对照组比较,RND1沉默组细胞增殖能力显著降低,凋亡率显著升高,侵袭细胞数和迁移细胞数明显减少,均P<0.05或<0.01.上皮标志蛋白E-钙黏蛋白表达上调,间质标志蛋白N-钙黏蛋白、波形蛋白及相关下游信号通路蛋白表达下调,均P<0.05.结论 RND1在TSCC中发挥促肿瘤作用,其沉默可通过抑制EMT过程减弱肿瘤细胞的增殖、侵袭和迁移能力,并促进凋亡.RND1具有潜在的治疗靶点价值.
Objective To evaluate the effects of RND1 silencing on the proliferation,apoptosis,invasion,and migration of tongue squamous cell carcinoma(TSCC)cells,and to explore the functional role of RND1 in TSCC progression.Methods RND1 protein expression levels in TSCC tissues and paired adjacent non-cancerous tissues were detected by Western blot and im-munohistochemistry.RND1 expression was silenced with siRNA in TSCC cell lines.Cell proliferation was assessed with the Cell Counting Kit-8(CCK-8)assay,apoptosis was evaluated with flow cytometry,and cell invasion and migration were assessed with Transwell and wound healing assays.In addition,Western blot analysis was performed to examine the expression of epithelial-mesenchymal transition(EMT)-related proteins and downstream signaling pathway-associated proteins.Results RND1 protein expression was significantly higher in TSCC tissues than in adjacent normal tissues(P<0.05).Compared with the blank and negative control groups,RND1-silenced cells exhibited significantly reduced proliferation,increased apoptosis,and markedly de-creased invasion and migration,with all differences being statistically significant(P<0.05 or<0.01).Meanwhile,the expres-sion of the epithelial marker E-cadherin was upregulated,whereas the expression of the mesenchymal markers N-cadherin and Vimentin,as well as downstream signaling pathway-related proteins,was downregulated(P<0.05).Conclusion RND1 plays a tumor-promoting role in TSCC.Silencing RND1 attenuates cell proliferation,invasion,and migration,and promotes apoptosis by suppressing the EMT process,suggesting that RND1 may serve as a potential therapeutic target for TSCC.
