外洗1号方治疗急性湿疹的作用机制探讨OA

Objective Exploring the treatment of acute eczema by external washing No.1 prescription based on net-work pharmacology technology Molecular Mechanisms.Methods:This study relied on the pharmacology database and analysis platform of traditional Chinese medicine system(TCMSP)and Pub Chem database to screen out the active in-gredients of 9 traditional Chinese medicines of Waixi No.1 prescription and predict the targets through the Swiss Target Prediction website;The disease targets of acute eczema were obtained by searching the human gene information data-base(Gene Cards),and the intersection genes between drug action targets and acute eczema targets were determined;The network diagram of drug active ingredients-action targets-diseases was constructed by Cytoscape3.10.3 software;Using STRING platform to construct protein interaction(PPI)network diagram for visual analysis;GO functional enrichment analysis and KEGG pathway analysis were performed through the DAVID website.Results:A total of 95 active ingredients were screened out from Waixi No.1 formula,corresponding to 438 targets,2513 targets for acute eczema,and 113 common targets for drugs and diseases.A total of 388 GO functional entries and 157 KEGG signaling pathways were obtained by enrichment analysis.Conclusion:The treatment of acute eczema by external washing No.1 formula may be through the action of IL6,TNF,IL1B,AKT1,and INS proteins,through regulating the positive regu-lation of MAPK cascade,inflammatory stimulation response,immune response,hypoxia response and other biological processes,mainly enriched in the pathways of PD-L1 expression and PD-1 checkpoint,biosynthesis and Phenylala-nine metabolism in cancer,correcting immune imbalance,inhibiting inflammation,and repairing skin barrier.
