多效唑对大豆籽粒蛋白质含量的影响及转录组分析OA

为明确大豆籽粒蛋白质含量响应多效唑的关键基因,本研究以宝豆1519为材料,以清水喷施为对照(CK),于花荚期每隔7 d喷施1次15%多效唑可湿性粉剂1 000倍稀释液,设置T1(喷施1次)、T2(喷施2次)和T3(喷施3次)3个处理,分析各处理籽粒蛋白质含量,并于鼓粒期取新鲜籽粒进行转录组学分析.结果表明,T2处理籽粒粗蛋白含量和水溶性蛋白含量最高,分别为46.11%和39.42%.各处理样品测序质量良好,有效reads占比在97.31%及以上,平均为97.68%,其中1 109个基因仅在T1表达,6 495个基因仅在T2表达,636个基因仅在T3表达,T2与CK的差异表达基因为8 451个.GO和KEGG富集分析表明,T2和CK的差异表达基因GO项主要富集在含氧化合物反应、水解酶活性负调控、对非生物刺激的反应和对脂质的反应等生物过程中;KEGG项主要富集在淀粉和蔗糖代谢、脂肪酸生物合成代谢、光合作用代谢等通路中.进一步分析表明,56个转录因子存在表达差异,NAC、ERF、C2H2和MYB等转录因子在多效唑的刺激下表达量提升,GLYMA_20G029600等10条差异表达上调基因与GLYMA_16G084000等10条差异表达下调基因可能与多效唑调节大豆籽粒蛋白质含量有关.本文为多效唑的合理施用提供参考.

To identify the key genes regulating soybean seed protein content in response to paclobutrazol,Baodou 1519 was used as the material in this study.A control group(CK)was sprayed with clean water.During the flowering and podding stage,a 1 000-fold diluted solution of 15%paclobutrazol wettable powder was sprayed every 7 days.3 treatment groups were established:T1(sprayed once),T2(sprayed twice),and T3(sprayed three times).The seed protein content of each treatment was analyzed,and fresh seeds were collected at the filling stage for transcriptomic analysis.The results showed that treatment T2 exhibited the highest crude protein content(46.11%)and water-soluble protein content(39.42%).Good sequencing quality was obtained for all treatment samples,with the proportion of valid reads exceeding 97.31%(average of 97.68%).Among the detected genes,1 109 genes were expressed exclusively in T1,6 495 genes exclusively in T2,and 636 genes exclusively in T3.A total of 8 451 differentially expressed genes(DEGs)were identified between the T2 and CK groups.GO and KEGG enrichment analyses indicated that the DEGs between T2 and CK were mainly enriched in biological processes such as response to oxygen-containing compounds,negative regulation of hydrolase activity,response to abiotic stimuli,and response to lipids.KEGG pathway analysis showed that the DEGs were primarily enriched in starch and sucrose metabolism,fatty acid metabolism,and photosynthesis metabolism.Further analysis revealed that 56 transcription factors were differentially expressed,and the expression levels of transcription factors such as NAC,ERF,C2H2,and MYB were found to be up-regulated in response to paclobutrazol stimulation.10 up-regulated DEGs(including GLYMA_20G029600)and 10 down-regulated DEGs(including GLYMA_16G084000)were identified as potentially involved in the regulation of soybean seed protein content by paclobutrazol.This study provides a reference for the rational application of paclobutrazol.
