目的:探究铜绿假单胞菌注射液(PA-MSHA)对健康供者免疫状态的影响,为提高亚健康人群免疫力提供理论依据.方法:于郑州大学第一附属医院招募15例健康供者并皮下注射PA-MSHA.采集注射PA-MSHA前及注射30 d后的外周血,流式细胞术检测各免疫细胞亚群比例、T细胞表面激活型与抑制型标志物表达及T细胞与NK细胞分泌促炎细胞因子的能力;高通量测序分析外周血T细胞受体(TCR)多样性.结果:接受PA-MSHA注射的志愿者外周血CD4/CD8升高;同时T细胞表面激活型标志物表达增加,而抑制型标志物表达降低;T细胞及NK细胞分泌促炎细胞因子如IFN-γ、Granzyme B和Perfo-rin的能力显著增强,且外周血TCR多样性增加.体外培养体系中,与对照组相比,PA-MSHA处理的CD3+T细胞功能显著增强.结论:PA-MSHA显著改善健康供者免疫状态,为亚健康人群提高免疫力提供了一种治疗方案.

Objective:To investigate impact of Pseudomonas aeruginosa-mannose sensitive hemagglutination(PA-MSHA)on immune status of healthy donors and to provide a theoretical foundation for enhancing immunity in sub-healthy individuals.Methods:A total of 15 healthy donors were recruited from the First Affiliated Hospital of Zhengzhou University and received subcutaneous injec-tions of PA-MSHA.Peripheral blood samples were collected prior to injection and 30 days post-injection,flow cytometric analysis was used to assess proportions of immune cell subsets,expressions of T cell surface activation and inhibition markers,as well as capacity of T cells and NK cells to secrete pro-inflammatory cytokines.High-throughput sequencing was employed to analyze T cell receptor(TCR)diversity.Results:Following PA-MSHA administration,there was a notable increase in CD4/CD8 among participants.Concur-rently,an elevation in activating marker expressions was observed alongside a reduction in inhibitory marker expressions on T cells.Furthermore,both T cells and NK cells exhibited significantly enhanced secretion capabilities for pro-inflammatory cytokines such as IFN-γ,Granzyme B and Perforin;additionally,an increase in peripheral blood TCR diversity was noted.In vitro experiments demon-strated that CD3+T cells treated with PA-MSHA displayed markedly improved functionality compared to control groups.Conclusion:PA-MSHA substantially enhances immune status of healthy donors and offers a viable therapeutic approach for bolstering immunity in sub-healthy populations.
