SIRT1/PGC-1α信号通路在糖尿病心肌病发生发展中的作用研究进展OA
Roles of SIRT1/PGC-1α signaling pathway in development and progression of diabetic cardiomyopathy:a review
糖尿病心肌病(DCM)是糖尿病特异性心脏并发症,其主要病理特征为心肌能量代谢紊乱与进行性心功能减退.近年研究发现,由沉默信息调节因子1(SIRT1)与过氧化物酶体增殖物激活受体γ辅激活因子1α(PGC-1α)构成的信号通路在DCM发病机制中扮演核心角色.本文系统综述SIRT1/PGC-1α信号通路的分子构成,重点探讨其通过调控线粒体质量控制、氧化应激平衡及能量代谢重编程参与DCM发生发展的最新研究进展,并对靶向该通路的天然活性成分、化学合成药物、基因治疗及其他干预策略进行评述,以期为DCM的防治提供新的理论依据与潜在治疗靶点.
Diabetic cardiomyopathy(DCM)is a diabetes-specific cardiac complication character-ized by myocardial metabolic dysfunction and progressive cardiac dysfunction.Recent studies have found that the signaling pathway formed by silent information regulator 1(SIRT1)and peroxisome proliferator-activated receptor γ coactivator 1 α(PGC-1α)plays a pivotal role in the pathogenesis of DCM.This review examines the molecular architecture of the SIRT1/PGC-1α signaling pathway in general and its involvement in DCM progression by regulating mitochondrial quality control,oxidative stress balance,and energy metabolism reprogramming in particular.This article also evaluates natural bioac-tive compounds,chemically synthesized drugs,gene therapies,and other intervention strategies targeting this pathway in the hope of providing a new line of thought and potential therapeutic targets for DCM prevention and treatment.
