目的 探讨蜂毒肽对骨关节炎(OA)大鼠软骨细胞凋亡及关节功能的影响,并分析其基于cGAS/STING信号通路的潜在机制.方法 将50只SD大鼠均分为5组:对照(Con)组、OA组、L-M组(0.5 mg/kg蜂毒肽腹腔注射)、H-M组(1 mg/kg蜂毒肽腹腔注射)和Vadimezan组(1 mg/kg蜂毒肽+25 mg/kg Vadimezan腹腔注射).通过评估关节肿胀度、功能评分,以及番红O-固绿和TUNEL染色观察软骨损伤与细胞凋亡,检测炎性细胞因子白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)和γ干扰素(IFN-γ)水平,并采用Western blotting分析细胞凋亡相关蛋白及cGAS/STING信号通路关键蛋白表达.结果 蜂毒肽可显著降低OA大鼠的关节肿胀度、功能评分、IL-1β、IL-6、IFN-γ水平及软骨细胞凋亡率,上调Bcl-2表达,下调Bax、caspase-3、cGAS和STING表达(P<0.05).结论 蜂毒肽可能通过抑制cGAS/STING信号通路,有效缓解OA大鼠的炎症反应及软骨细胞凋亡,进而减轻软骨组织损伤并改善关节功能.

Objective To explore the effect of melittin on chondrocyte apoptosis and joint function in rats with osteoarthritis(OA)and analyze its potential mechanism based on the cGAS/STING signaling pathway.Methods Fifty SD rats were evenly divided into five groups:Con(control),OA,L-M(0.5 mg/kg melittin intraperitoneal injection),H-M(1 mg/kg melittin intraperitoneal injection),and Vadi-mezan(1 mg/kg melittin+25 mg/kg Vadimezan intraperitoneal injection).The degree of joint swelling and the functional scores were evaluated.Cartilage damage and chondrocyte apoptosis were observed using Safranin O-Fast Green and TUNEL staining.The levels of inflammatory cytokines IL-1β,IL-6,and IFN-γ were detected.Western blotting was used in order to analyze the expression of apoptosis-re-lated proteins and key proteins in the cGAS/STING signaling pathway.Results Melittin significantly reduced the joint swelling degree,functional scores,inflammatory cytokine levels,and chondrocyte apoptosis rate in OA rats.It upregulated Bcl-2 expression and downre-gulated Bax,caspase-3,cGAS,and STING expression(P<0.05).Conclusion Melittin may effectively alleviate inflammatory responses and chondrocyte apoptosis in rats with OA by inhibiting the cGAS/STING signaling pathway,thereby reducing cartilage tissue damage and improving joint function.
