目的 探究花椒麻素调节AMP活化蛋白激酶(AMPK)/乙酰辅酶A羧化酶(ACC)/固醇调节元件结合蛋白1(SREBP1)信号通路对妊娠期糖尿病(GDM)大鼠糖脂代谢、氧化应激的影响.方法 将SD大鼠随机分为对照组、模型组、低剂量花椒麻素组、中剂量花椒麻素组、高剂量花椒麻素组、联合组,每组12只,检测各组大鼠血糖、血脂、脂肪因子和氧化应激指标水平.采用Western blotting检测AMPK/ACC/SREBP1信号通路相关蛋白表达.结果 与对照组相比,模型组空腹血糖(FPG)、空腹胰岛素(FINS)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、瘦素、丙二醛(MDA)水平以及SREBP1表达水平升高,高密度脂蛋白胆固醇(HDL-C)、脂联素(APN)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)水平以及磷酸化AMPKα(p-AMPKα)/AMPKα、磷酸化ACC(p-ACC)/ACC表达水平降低(P<0.05).与模型组相比,低、中、高剂量花椒麻素组FPG、FINS、TC、TG、LDL-C、瘦素、MDA水平以及SREBP1表达水平降低,HDL-C、APN、SOD、GSH-Px水平以及p-AMPKα/AMPKα、p-ACC/ACC表达水平升高(P<0.05).与高剂量花椒麻素组相比,联合组FPG、FINS、TC、TG、LDL-C、瘦素、MDA水平以及SREBP1表达水平升高,HDL-C、APN、SOD、GSH-Px水平以及p-AMPKα/AMPKα、p-ACC/ACC表达水平降低(P<0.05).结论 花椒麻素可能通过激活AMPK/ACC/SREBP1信号通路,抑制氧化应激,进而改善GDM大鼠的糖脂代谢水平.

Objective To investigate the effects of sanshool on glycolipid metabolism and oxidative stress in rats with gestational dia-betes mellitus(GDM)via the AMPK/ACC/SREBP1 signaling pathway.Methods SD rats were randomly divided into control,model,low-dose sanshool,medium-dose sanshool,high-dose sanshool,and combined groups,with 12 rats in each group.Blood glucose,blood lipids,adipokines,and oxidative stress indexes were measured.Expressions of proteins related to the AMPK/ACC/SREBP1 signaling pathway were detected using Western blotting.Results Compared with the control group,the model group showed increased levels of fasting plasma glucose(FPG),fasting insulin(FINS),total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),leptin,malondialdehyde(MDA),and SREBP1,and decreased levels of high-density lipoprotein cholesterol(HDL-C),adiponectin(APN),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),p-AMPKα/AMPKα,and p-ACC/ACC(P<0.05).Compared with the model group,the low-,medium-,and high-dose sanshool groups showed decreased levels of FPG,FINS,TC,TG,LDL-C,leptin,MDA,and SREBP1,and increased levels of HDL-C,APN,SOD,GSH-Px,p-AMPKα/AMPKα,and p-ACC/ACC(P<0.05).Compared with the high-dose sanshool group,the combined group showed increased levels of FPG,FINS,TC,TG,LDL-C,leptin,MDA,and SREBP1,and decreased levels of HDL-C,APN,SOD,GSH-Px,p-AMPKα/AMPKα,and p-ACC/ACC(P<0.05).Conclusion Sanshool may improve glycolipid metabolism and inhibit oxidative stress in GDM rats,potentially through activation of the AMPK/ACC/SREBP1 signaling pathway.
