野马追中化学成分及其肺保护作用研究OA
Chemical constituents of Eupatorii Lindleyani Herba and their lung-protective effects
该研究旨在探讨野马追的化学成分,并筛选具有肺保护作用的活性成分.采用硅胶吸附柱色谱、ODS 柱色谱及HPLC 等多种色谱方法对野马追石油醚萃取物进行分离纯化,根据各化合物的理化性质及波谱数据进行结构鉴定.结果共分离得到9 个化合物,分别鉴定为eupaphenolide A(1)、eupaphenolide B(2)、aurantiamide acetate(3)、aurantiamide(4)、8β-tigloiloxi-grazielia(5)、(3S,6R,7R,8R)-3-hydroxy-8-acetoxysarracenyloxy germacra-1(10),4,11(13)-trien-6,12-ilide(6)、eucannabinolide(7)、eupalinolide G(8)、eupachinsin A(9),其中化合物1 和2 为新化合物.经MTT 法评价其体外肺保护活性,结果显示吉马烷型倍半萜类化合物(5~9)均表现出一定的肺保护作用,其中化合物 8 在 1 μmol·L-1浓度下显著提高细胞存活率,作用强于阳性对照甘草查尔酮 A,而化合物 6、7、9 在较低浓度下亦能部分改善 NNK 诱导的细胞损伤,呈现低浓度保护、高浓度抑制的双相效应.研究表明,野马追所含吉马烷型倍半萜类成分可能是其肺保护活性的主要物质基础,为野马追的药效物质研究提供了实验依据.
This study aimed to investigate the chemical constituents of Eupatorii Lindleyani Herba and to identify the compounds with protective effects on the lung.The petroleum ether extract of Eupatorii Lindleyani Herba was separated and purified through silica gel column chromatography,ODS column chromatography,and HPLC.The structures of the isolated compounds were identified based on their physicochemical properties and spectroscopic data.Nine compounds were obtained and identified as eupaphenolide A(1),eupaphenolide B(2),aurantiamide acetate(3),aurantiamide(4),8β-tigloiloxi-grazielia(5),(3S,6R,7R,8R)-3-hydroxy-8-acetoxysarracenyloxy germacra-1(10),4,11(13)-trien-6,12-ilide(6),eucannabinolide(7),eupalinolide G(8),and eupachinsin A(9).Compounds 1 and 2 were newly identified.The lung-protective activities of these compounds were evaluated in vitro via the MTT assay.Germacranolide-type sesquiterpenes(5-9)showed notable protective effects,among which compound 8 at 1 μmol·L-1 significantly increased cell viability,exhibiting stronger activity than the positive control licochalcone A.Compounds 6,7,and 9 at low concentrations partially reduced NNK-induced cell injury and displayed a biphasic effect characterized by protection at low concentrations and inhibition at high concentrations.These results suggest that the germacranolide-type sesquiterpenes in Eupatorii Lindleyani Herba may serve as the main active constituents responsible for its lung-protective effects,providing experimental evidence for further pharmacological research on this plant.
