两个香菇品种贮藏特性及蛋白组学的比较分析OA

为探究香菇耐贮性的生物学机制并筛选耐贮相关蛋白,以耐贮品种豫香 1 号(YX)和不耐贮品种 9608(JL)为试材,系统比较二者贮藏特性、细胞膜稳定性、抗氧化能力、细胞壁组分及糖代谢等生理指标,并结合蛋白组学技术开展差异分析.结果表明,贮藏期间,YX 始终维持较低的失水率、褐变度和呼吸速率,硬度下降平缓;其相对电导率、丙二醛(MDA)含量显著低于 JL,超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性保持在较高水平.在细胞壁组分方面,YX 的纤维素、几丁质含量更高且降解缓慢,而纤维素酶、几丁质酶活性显著低于 JL;在糖代谢层面,YX 可溶性糖总含量降幅更小,海藻糖等关键组分含量明显高于 JL.蛋白组学分析共鉴定到 5474 个蛋白,YX 贮藏前后的差异表达蛋白(95 个)远少于 JL(789 个),表明耐贮品种蛋白表达稳定性更强.进一步筛选出葡聚糖酶(LENED_012880)和胍丁胺酶(LENED_003685)为耐贮相关候选蛋白,二者在 YX 贮藏过程中呈显著上调表达.GO 和KEGG 富集分析显示,该耐贮相关蛋白主要参与细胞壁组织、甘油磷脂代谢、淀粉和蔗糖代谢等通路.综上,YX 通过协同维持低代谢水平、强化抗氧化防御体系及稳定细胞壁结构,实现了优异的耐贮性.本研究筛选的耐贮相关蛋白为香菇耐贮品种分子育种提供了核心靶点,相关研究结果也为解析香菇采后衰老调控机制及优化保鲜技术提供了理论支撑.

To explore the biological mechanisms underlying the storability of Lentinula edodes and screen key proteins as-sociated with storability,this study employed the storage-tolerant variety Yuxiang 1(YX)and the storage-intolerant vari-ety 9608(JL)as test materials.A systematic comparison was conducted on their physiological indicators including storage characteristics,cell membrane stability,antioxidant capacity,cell wall components,and sugar metabolism,combined with proteomic techniques for differential analysis.The results showed that during storage,YX consistently maintained low wa-ter loss rate,browning degree,and respiratory rate,with a slow decrease in firmness.Moreover,its relative electrical con-ductivity and malondialdehyde(MDA)content were significantly lower than those of JL,while the activity of superoxide dismutase(SOD)and catalase(CAT)remained at a high level.Regarding cell wall components,YX had higher content of cellulose and chitin which degraded slowly,whereas the activity of cellulase and chitinase were significantly lower than those of JL.In terms of sugar metabolism,the total soluble sugar content of YX decreased less significantly,and the con-tent of key components such as trehalose were significantly higher than those of JL.Proteomic analysis identified a total of 5474 proteins,and the number of differentially expressed proteins(DEPs)in YX before and after storage(95)was much fewer than that in JL(789),indicating stronger stability of protein expression in the storage-tolerant variety.Further screening revealed that glucanase(LENED_012880)and agmatinase(LENED_003685)were key storability-related pro-teins,both of which were significantly up-regulated during the storage of YX.GO and KEGG enrichment analysis demon-strated that these key proteins were mainly involved in pathways such as cell wall organization,glycerophospholipid me-tabolism,and starch and sucrose metabolism.In conclusion,YX achieves excellent storability through the synergistic ef-fects of maintaining a low metabolic level,enhancing the antioxidant defense system,and stabilizing cell wall structure.The key storability-related proteins screened in this study provide core targets for the molecular breeding of storage-toler-ant L.edodes varieties,and the relevant results also offer theoretical support for deciphering the regulatory mechanisms of postharvest senescence in L.edodesand optimizing preservation technologies.
