探讨生、炙黄芪中总皂苷、总黄酮、总多糖对脾虚大鼠肠道菌群、短链脂肪酸代谢和免疫活性的影响.采用饮食不节+苦寒泻下+劳倦过度相结合的方法建立脾虚模型,16S rRNA 高通量测序技术分析生、炙黄芪不同组分对脾虚大鼠肠道菌群结构多样性的调控作用,顶空气相色谱-质谱联用技术测定生、炙黄芪不同组分对脾虚大鼠短链脂肪酸代谢的调节作用,ELISA 技术评价生、炙黄芪对脾虚大鼠特异性和非特异性免疫活性的影响作用.结果表明,生、炙黄芪的总黄酮、总皂苷、总多糖可明显降低脾虚大鼠肠道中有害菌属螺杆菌属的丰度,同时除生、炙黄芪总多糖外,其余各给药组亦均可明显降低有害菌属萨特菌属的丰度(P<0.05).生、炙黄芪中各组分能够回调脾虚大鼠结肠中各种短链脂肪酸的水平,其中炙黄芪总多糖提升乙酸含量的作用明显优于生黄芪总多糖(P<0.05).生、炙黄芪的总多糖均可显著回调脾虚大鼠血清和结肠中 IL-4、IL-6、IL-10、IL-18、IgA、IgG 的水平以及血清中 IL-2的水平(P<0.05,P<0.01),且炙品回调作用优于生品.综上,生黄芪经蜜炙后各组分特异性免疫和非特异性免疫调节作用增强,从而增强了改善脾虚的作用.这可为阐释黄芪经蜜炙能够增强益气补中作用的科学内涵提供参考依据.

This study aimed to investigate the effects of total saponins,total flavonoids,and total polysaccharides from raw and honey-processed Astragali Radix on intestinal microbiota,short-chain fatty acid metabolism,and immune activity in rats with spleen deficiency.A spleen deficiency model was established using a combination of irregular diet,bitter-cold purgative administration and excessive fatigue.High-throughput 16S rRNA sequencing was employed to analyze the modulatory effects of different fractions from raw and honey-processed Astragali Radix on the structural diversity of intestinal microbiota in spleen-deficient rats.Headspace gas chromatography-mass spectrometry was used to determine the modulatory effects of these fractions on the metabolism of short-chain fatty acids.ELISA was employed to assess the effects of raw and honey-processed Astragali Radix on specific and non-specific immune activities of spleen-deficient rats.The results demonstrated that the total flavonoids,total saponins,and total polysaccharides from raw and honey-processed Astragali Radix significantly restored the abundance of the harmful bacterial genus Helicobacter in the intestines of spleen-deficient rats.Moreover,except for the total polysaccharides from both raw and honey-processed Astragali Radix,all other treatment groups also significantly reduced the abundance of the harmful bacterial genus Sutterella(P<0.05).The fractions from raw and honey-processed Astragali Radix were able to restore the levels of various short-chain fatty acids in the colon of spleen-deficient rats.Notably,the total polysaccharides from honey-processed Astragali Radix significantly outperformed those from raw Astragali Radix in increasing acetic acid content(P<0.05).The total polysaccharides of both raw and honey-processed Astragali Radix significantly restored the levels of IL-4,IL-6,IL-10,IL-18,IgA,and IgG in the serum and colon,as well as the level of IL-2 in the serum of spleen-deficient rats(P<0.05 or P<0.01).The restorative effects of the processed product were superior to those of the raw product.In summary,the specific and non-specific immunomodulatory effects of the components from raw Astragali Radix increased after honey-processing,thereby potentiating its efficacy in ameliorating spleen deficiency.These findings may provide a scientific basis for elucidating the mechanism by which honey-processing enhances the qi-tonifying and middle-strengthening effects of Astragali Radix.
