目的 基于数据挖掘与网络药理学技术,探讨含百合中成药治疗失眠的用药规律与潜在作用机制.方法 通过药智网收集并筛选含百合中成药处方,借助SPSS软件进行用药频次、性味归经、关联规则及聚类分析,确定核心药组.利用BAT-MAN-TCM、GeneCards等数据库筛选核心药组的活性成分、疾病靶点及交集靶点,进一步通过STRING、DAVID完成蛋白质-蛋白质相互作用(PPI)网络构建与通路富集分析.结果 共纳入 73 首含百合中成药,其中以失眠为主症的心系疾病处方 17 首.高频药物前 5 位分别为百合、茯苓、酸枣仁、远志、当归.关联规则显示"百合-茯苓"支持度最高(64.706%),确定其为核心药对.根据高频药物及聚类分析,以"百合-茯苓"为核心,纳入酸枣仁、远志、当归,构建核心药组.核心药组共筛选出 184 种核心活性成分,与失眠交集得 454 个共同靶点;PPI网络分析筛选出 81 个核心靶点,如肿瘤蛋白 53(TP53)、蛋白激酶B1(AKT1)等.京都基因与基因组百科全书(KEGG)富集主要涉及磷脂酰肌醇 3 激酶-蛋白激酶 B(PI3K-AKT)、丝裂原活化蛋白激酶(MAPK)等信号通路.结论 含百合中成药治疗失眠以"百合-茯苓"为核心药对,其机制可能通过姜黄素、大黄素等多成分作用于TP53、AKT1 等多个靶点,调控PI3K-AKT等多条信号通路,经由调节神经内分泌、抗炎、抗氧化等途径实现对失眠的干预作用.

Objective To explore the medication rules and potential mechanism of action of Chinese patent medicines containing Baihe(Lilii Bulbus)in the treatment of insomnia based on data mining and network pharmacology.Methods Prescriptions of Chinese patent medicines containing Baihe(Lilii Bulbus)were collected and screened through the Yaozhi network.SPSS software was used to perform analyses of medication frequency,properties,flavors,and meridian tropism,association rules,and cluster analysis to identify the core drug combination.Databases including BATMAN-TCM and GeneCards were used to screen the active components of the core drug combinations,disease targets,and intersecting targets.STRING and DAVID were further employed to construct the protein-protein interaction(PPI)network and conduct pathway enrichment analysis.Results A total of 73 prescriptions of Chinese patent medicines containing Baihe(Lilii Bulbus)were included,among which 17 were for heart system diseases with insomnia as the primary symptom.The top five high-frequency drugs were Baihe(Lilii Bulbus),Fuling(Poria),Suanzaoren(Ziziphi Spinosae Semen),Yuanzhi(Polygalae Radix),and Danggui(Angelicae Sinensis Radix).Association rule analysis showed that the"Baihe(Lilii Bulbus)-Fuling(Poria)"pair had the highest support(64.706%),and it was identified as the core drug pair.Based on high-frequency drugs and cluster analysis,a core drug combination was constructed with"Baihe(Lilii Bulbus)-Fuling(Poria)"as the core,incorporating Suanzaoren(Ziziphi Spinosae Semen),Yuanzhi(Polygalae Radix),and Danggui(Angelicae Sinensis Radix).A total of 184 core active components were screened from the core drug combination,yielding 454 intersecting targets with insomnia.PPI network analysis screened out 81 core targets,including tumor protein 53(TP53)and protein kinase B1(AKT1).Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis mainly involved signaling pathways such as phosphatidylinositol 3-kinase-protein kinase B(PI3K-AKT)and mitogen-activated protein kinase(MAPK).Conclusion The treatment of insomnia with Chinese patent medicines containing Baihe(Lilii Bulbus)takes"Baihe(Lilii Bulbus)-Fuling(Poria)"as the core drug pair.The mechanism may involve multiple components such as curcumin and emodin acting on multiple targets including TP53 and AKT1,regulating multiple signaling pathways such as PI3K-AKT,and exerting intervention effects on insomnia through neuroendocrine regulation,anti-inflammation,and antioxidant pathways.
