铁死亡是由 Fe2+超载、氨基酸代谢失常、脂质过氧化物累积诱发的新型程序性细胞死亡.炎症是机体对感染、损伤或其他刺激的一种防御机制.炎症反应能激活多种炎症相关信号通路,通过调节铁代谢、ROS产生、抗氧化防御、脂质代谢和线粒体功能等多个方面共同促发铁死亡.在炎症相关的信号转导通路中,本文重点关注了JAK-STAT、NF-κB、炎症体、c GAS-STING和MAPK这5条经典的炎症通路,阐述它们在铁死亡中的作用,有助于进一步了解铁死亡的病理生理过程.通过研究这些与炎症相关的通路,为更深入探究铁死亡的机制提供新的思考和研究方向,为干预铁死亡相关炎症疾病提供合适的治疗途径.

Ferroptosis is a new type of programmed cell death induced by Fe2+overload,abnormal amino acid metabolism and lipid peroxide accumulation.Inflammation is the body's defense mechanism against infection,injury or other irritation.Inflammatory response can activate a variety of inflammation-related signaling pathways and jointly promote the occurrence of ferroptosis by regulat-ing iron metabolism,ROS production,antioxidant defense,lipid metabolism and mitochondrial function.Among the relevant signal transduction pathways,this paper focuses on five classical inflammatory pathways,namely,JAX-STAT,NF-κB,inflammasome,c GAS-STING and MAPK and elaborates their roles in ferroptosis,which is helpful to further understand the pathophysical process of iron death.Study of these inflammatory pathways provides new thinking and research directions for further exploring the mechanism of ferroptosis and provides appropriate therapeutic approaches for the intervention of ferroptosis-related diseases.
