目的:观察电针通过环磷酸鸟苷-腺苷合成酶(cGAS)-干扰素基因刺激因子(STING)信号通路调节巨噬细胞极化对急性胃黏膜损伤(AGMI)小鼠的影响.方法:将C57BL/6小鼠随机分为正常组、模型组和电针组,每组 10只.以无水乙醇灌胃构建 AGMI模型.电针组小鼠给予"中脘""足三里"电针治疗,30 min/次,1次/d,共5 d.采用 Guth 胃黏膜损伤评分法对小鼠的胃黏膜损伤进行评估;HE染色和PAS染色观察小鼠胃黏膜组织病理变化;流式细胞术分析小鼠胃黏膜组织M1、M2型巨噬细胞比例;实时荧光定量PCR法检测小鼠胃黏膜组织肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)、转化生长因子-β(TGF-β)mRNA 表达水平;Western blot法检测小鼠胃黏膜组织闭合小环蛋白 1(ZO-1)、闭合蛋白(Occludin)、cGAS、STING、磷酸化 STING(p-STING)、磷酸化干扰素调节因子 3(p-IRF3)、磷酸化TANK 结合激酶 1(p-TBK1)、诱导型一氧化氮合酶(iNOS)、分化簇 86(CD86)、精氨酸酶 1(Arg-1)、分化簇 206(CD206)的蛋白表达水平;免疫荧光检测胃黏膜组织 iNOS、CD206荧光强度.结果:与正常组相比,模型组小鼠胃黏膜损伤指数升高(P<0.01),胃黏膜上皮坏死、腺体紊乱、黏液层破坏,胃黏膜组织M1型巨噬细胞增多、M2型巨噬细胞减少(P<0.01),TNF-α、IL-6 的 mRNA水平升高(P<0.01),IL-10、TGF-β的 mRNA水平降低(P<0.01),ZO-1、Occludin蛋白表达显著降低(P<0.01),cGAS-STING通路相关蛋白及iNOS、CD86上调(P<0.01),Arg-1、CD206下调(P<0.01).与模型组比较,电针组上述指标均有所逆转(P<0.01,P<0.05),胃黏膜病理损伤减轻,黏液层较完整.结论:电针可减轻胃黏膜炎性反应,改善AGMI,其作用机制可能与抑制cGAS-STING信号通路介导的MI型巨噬细胞极化有关.

Objective To observe the effect of electroacupuncture on mice with acute gastric mucosal injury(AGMI)by regulating macrophage polarization through the cyclic guanosine monophosphate-adenosine synthase(cGAS)-stimulator of interferon genes(STING)signaling pathway.Methods Thirty C57BL/6 mice were randomly divided into normal group,model group,and electroacupuncture group.Except for the normal group,the remaining mice were intragastrically administered with absolute ethanol to establish the AGMI model.Mice in the electroacupuncture group were treated with electroacupuncture at"Zhongwan"(CV12)and"Zusanli"(ST36)for 30 min,once a day for 5 consecutive days.The Guth gastric mucosal injury scoring method was used to evaluate gastric mucosal injury.HE staining and PAS staining were performed to observe the pathological changes of gastric mucosal tissue.Flow cytometry was used to analyze the proportions of M1 and M2 macrophages in gastric mucosal tissue.Real-time quantitative PCR was used to detect the mRNA expression levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-10(IL-10),and transforming growth factor-β(TGF-β)in gastric mucosal tissue.Western blot was used to detect the protein expression levels of zonula occludens-1(ZO-1),Occludin,cGAS,STING,phosphorylated STING(p-STING),phosphorylated interferon regulatory factor 3(p-IRF3),phosphorylated TANK-binding kinase 1(p-TBK1),inducible nitric oxide synthase(iNOS),cluster of differentiation 86(CD86),arginase-1(Arg-1),and cluster of differentiation 206(CD206).Immunofluorescence was used to detect the fluorescence intensity of iNOS and CD206 in gastric mucosal tissue.Results Compared with the normal group,the model group showed an increased gastric mucosal injury index(P<0.01),with epithelial necrosis,glandular disorder,and mucus layer destruction in the gastric mucosa;the number of M1 macrophages increased while M2 macrophages decreased(P<0.01);the mRNA expression levels of TNF-α and IL-6 were increased(P<0.01),while the mRNA expression levels of IL-10 and TGF-β were decreased(P<0.01);the protein expression levels of ZO-1 and Occludin were significantly decreased(P<0.01);the proteins related to the cGAS-STING pathway,as well as iNOS and CD86 were up-regulated(P<0.01),while Arg-1 and CD206 were down-regulated(P<0.01)in gastric mucosal tissue.Compared with the model group,all the above mentioned indicators in the electroacupuncture group were significantly reversed(P<0.01,P<0.05),with reduced pathological damage and a relatively intact mucus layer in gastric mucosa.Conclusion Electroacupuncture can alleviate gastric mucosal inflammation and improve AGMI,and its mechanism may be related to inhibiting MI macrophage polarization mediated by the cGAS-STING signaling pathway.
