Research Progress on the Relationship Between VEGF and Liver Fibrosis
肝纤维化是肝脏对肝损伤的修复过程,是各种慢性肝病进展过程中肝内纤维生成与降解失衡,致使过多的胶原在肝内沉积的结果.肝纤维化可发展为肝硬化,严重威胁人类生命健康.血管内皮生长因子(VEGF)家族是一类多功能的细胞因子,在肝脏保护和再生、血管及淋巴管的生成过程中起着重要的作用,可促进内皮细胞增殖、促进血管生成以及增加血管的通透性.研究表明,VEGF是致肝纤维化发生的重要因素之一,近年来国内外对VEGF在肝纤维化中作用的研究取得了一定的进展,为肝纤维化发病机制的阐明及临床药物的研发提供了新的理论依据.本文拟对VEGF在诱导HSCs激活、肝脏缺氧状态下、炎症过程中、肝窦内皮细胞中导致肝纤维化的作用机制进行总结,以期为抗纤维化的科学研究及临床治疗提供理论依据.
Liver fibrosis is a repair process of the liver in response to liver injury and is the result of an imbalance between intrahepatic fibrogenesis and fibrolysis during the progression of various chronic liver diseases,leading to excessive collagen deposition within the liver.Liver fibrosis can progress to cirrhosis,seriously threatening human life and health.Vascular endothelial growth factor(VEGF)family is a group of multifunctional cytokines that play important roles in liver protection and regeneration,as well as in angiogenesis and lymphangiogenesis,and can promote endothelial cell proliferation,promote angiogenesis,and increase vascular permeability.Studies have shown that VEGF is one of the important factors leading to the development of liver fibrosis.In recent years,research on the role of VEGF in liver fibrosis has made certain progress both domestically and internationally,providing new theoretical basis for elucidating the pathogenesis of liver fibrosis and for the development of clinical drugs.This article aims to summarize the mechanisms of action by which VEGF induces liver fibrosis in the contexts of inducing HSCs activation,under hepatic hypoxia,during inflammation,and in liver sinusoidal endothelial cells,in order to provide a theoretical basis for scientific research and clinical treatment of anti-fibrosis.
