Quality grading standards of Gleditsia sinensis seeds based on the correlation between phenotypic traits and polysaccharide content

目的 制定贵州、山东及河南等主产区皂角米的质量分级标准.方法 分别对皂角米的长度、宽度、长宽比、厚度、百粒重、色度、黏度、膨胀度和多糖含量进行测定,采用Pearson相关性分析判断不同性状与成分间的相关性,根据相关性分析、主成分分析及K-均值聚类分析的结果得出划分等级的主要指标,并制定出皂角米质量的分级标准.结果 皂角米形态-多糖含量相关性分析结果表明皂角米的长度、宽度、厚度、百粒重都与多糖含量呈极显著性正相关;多糖含量与色度值L*存在极显著正相关关系,而多糖含量与色度值a*存在极显著负相关关系.基于K-均值聚类,拟定三级标准:Ⅰ级(长度≥11.38 mm、宽度≥7.34 mm、百粒重≥23.71 g、多糖含量≥73.67%)、Ⅱ级(长度 10.68~11.38 mm、百粒重 18.18~23.71 g、多糖含量 70.45%~73.67%)、Ⅲ级(长度≤10.68 mm、百粒重≤18.18 g、多糖含量≤70.45%).二级标准以Ⅱ级与Ⅲ级的分界值为下限,将原Ⅰ级和Ⅱ级合并为新的Ⅰ级,原Ⅲ级作为新Ⅱ级.结论 以贵州、山东及河南等主产区皂角米为研究对象,通过表型性状与多糖含量的关联分析,初步构建了皂角米质量分级标准,揭示了表观性状与多糖含量的内在联系,可为产区优质种质筛选及标准化生产提供参考.

Objective To establish a quality grading standard for Gleditsia sinensis seed produced in major producing areas including Guizhou,Shandong,and Henan Provinces.Methods The length,width,length-width ratio,thickness,100-kernel weight,chromaticity,viscosity,swelling capacity,and polysaccharide content of Gleditsia sinensis seed were determined.Pearson correlation analysis was adopted to analyze the correlation among different traits and components.Based on the results of correlation analysis,principal component analysis,and K-means cluster analysis,the key indicators for grading were determined,and the quality grading standard for Gleditsia sinensis seed was formulated.Results The correlation analysis between morphological traits and polysaccharide content of Gleditsia sinensis seeds showed that the length,width,thickness,and 100-kernel weight of Gleditsia sinensis seeds had extremely significant positive correlations with polysaccharide content.Polysaccharide content exhibited an extremely significant positive correlation with chromaticity value L*,and an extremely significant negative correlation with chromaticity value a*.Based on K means clustering,a three grade standard was proposed:Grade I(length≥11.38 mm,width≥7.34 mm,100 kernel weight≥23.71 g,polysaccharide content≥73.67%),Grade II(length 10.68-11.38 mm,100 kernel weight 18.18-23.71 g,polysaccharide content 70.45%-73.67%),and Grade III(length≤10.68 mm,100 kernel weight≤18.18 g,polysaccharide content≤70.45%).A two grade standard was then defined by using the cutoff values between Grade II and Grade III as the lower limit,merging the original Grade I and Grade II into the new Grade I,and retaining the original Grade III as the new Grade II.Conclusion Taking Gleditsia sinensis seeds from major producing areas of Guizhou,Shandong,and Henan as the research subjects,a preliminary quality grading standard for Gleditsia sinensis seeds was established through correlation analysis between phenotypic traits and polysaccharide content.The intrinsic relationship between apparent traits and polysaccharide content was revealed,providing a reference for high quality germplasm screening and standardized production in these regions.
