根皮苷、三叶苷与根皮素的体外稳定性研究OA
In Vitro Stability Studies of Phlorizin,Trilobatin,and Phloretin
目的 探究根皮苷、三叶苷以及根皮素的体外稳定性.方法 采用液质联用考察根皮苷、三叶苷及根皮素在不同pH(1.4、4.0、6.86、7.86)缓冲液、人工模拟胃液、肠液及大鼠胃、肠内容物中的稳定性.结果 根皮苷与三叶苷在强酸性环境中降解迅速,三叶苷降解速率高于根皮苷;在中性环境中,二者降解减缓;在弱碱性环境中,初始降解迅速,随后趋于平缓;在人工胃、肠液及大鼠胃、肠内容物中,二者降解速率更快且伴随根皮素的生成,而根皮素在胃酸及肠液中均降解迅速,半衰期极短.结论 根皮苷与三叶苷在胃肠道环境中稳定性差,易水解生成根皮素,且根皮素自身又极不稳定,严重影响其生物利用度.
Objective To investigate the in vitro stability of phlorizin,trilobatin,and phloretin.Methods High performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS)was employed to evaluate the stability of phloridzin,trilobatin,and phloretin in buffer solutions of various pH values(1.4,4.0,6.86,and 7.86),simulated gastric and intestinal fluids,and rat gastric and intestinal con-tents.Results Phlorizin and trilobatin degraded rapidly in strong acidic conditions,with trilobatin degrading at a higher rate than phlorizin.In neutral environments,the degradation of both compounds slows down.Under weakly alkaline conditions,initial degradation was rapid,but subsequently levels off.In both artificial and rat gastrointestinal fluids,the degradation of phlorizin and trilobatin were accelerated and ac-companied by the generation of phloretin.However,phloretin itself degraded rapidly in gastric acid and intestinal fluid,possessing an ex-tremely short half-life.Conclusion Phlorizin and trilobatin possess poor stability in the gastrointestinal environment,where they are prone to hydrolysis into phloretin.Furthermore,since phloretin itself is highly unstable,this instability significantly compromises their bioavailability.
