目的 探究中药百合水提物对 SH-SY5Y 细胞生长的影响以及对皮质酮诱导 SH-SY5Y 细胞损伤的神经保护作用.方法 体外培养的 SH-SY5Y 细胞与不同浓度的百合水提物作用 8h 后,加入 10μmol/L 皮质酮作用 24h,诱导 SH-SY5Y 细胞损伤.采用 MTT 法测定细胞存活率,并进一步采用免疫印迹检测脑源性神经生长因子(BDNF)和神经生长因子(NGF)的蛋白水平.结果 百合水提物在6.25~800μg/mL 条件下对正常 SH-SY5Y 细胞不产生毒性作用;10μmol/L皮质酮处理后,细胞存活率显著降低,而百合水提物在 100、200μg/mL 浓度下预保护 8h 可显著提高细胞存活率.皮质酮损伤后的 SH-SY5Y 细胞 BDNF 和 NGF 蛋白水平均显著下调,百合水提物(50、100、200μg/mL)干预后其表达水平呈现剂量依赖性上调.结论百合水提物对皮质酮损伤的 SH-SY5Y 细胞具有明显的保护作用,其机制可能与其调控神经营养因子信号通路密切相关.

Objective To investigate the effects of an aqueous extract of Lilii Bulbus(AELB)on the growth of SH-SY5 Y cells,and its neuroprotective role against corticosterone(CORT)-induced neuronal injury in these cells.Methods SH-SY5 Y cells were pretreated with various concentrations of AELB for 8h,followed by exposure to 10μmol/L CORT for 24h to establish an in vitro neuronal injury model.Cell viability was assessed using the MTT assay,and the protein levels of brain-derived neurotrophic factor(BDNF)and nerve growth factor(NGF)were measured by Western blotting.Results AELB(6.25~800μg/mL)exhibited no cytotoxicity toward normal SH-SY5 Y cells.CORT(10μmol/L)significantly reduced cell viability,whereas pretreatment with AELB(100 and 200μg/mL)for 8h markedly improved cell viability.Both BDNF and NGF protein levels were significantly downregulated in CORT-injured cells but were dose-dependently restored by AELB(50,100,and 200μg/mL).Conclusion AELB demonstrates notable neuroprotective effects against CORT-induced SH-SY5 Y cell injury,and its underlying mechanism may be closely associated with its regulation of neurotrophic factor signaling pathways.
