貂源肺炎克雷伯菌毒力基因及耐药特征分析OA
Analysis of Virulence Genes and Drug Resistance Characteristics of Klebsiella pneumoniae Isolated from Mink
肺炎克雷伯菌主要引起水貂肺炎,严重影响水貂养殖业的发展.本研究主要分析貂源肺炎克雷伯菌的毒力和耐药情况,为研究貂源肺炎克雷伯菌的致病性和耐药性提供理论依据.本研究通过细菌纯培养、革兰氏染色、PCR方法对菌株进行分离鉴定;采用琼脂稀释法检测菌株的耐药谱,PCR方法鉴定菌株的血清型.提取菌株全基因组DNA并进行测序,分析菌株全基因组序列的组成以及重要成分.利用全基因组序列对菌株的基因型、耐药基因及毒力基因进行了生物信息学分析.结果表明,分离菌株为肺炎克雷伯菌,该菌株属K2血清型.药敏试验结果显示,菌株对四环素、氯霉素、氟苯尼考、氨苄西林、头孢他啶、头孢西丁、环丙沙星、恩诺沙星和卡那霉素表现为耐药性,呈现多重耐药特征.组装后的基因组大小为5 435.522 kb,含有一个染色体和一个质粒.全基因组分析表明菌株属于ST86型.经过比对分析表明,菌株携带7种毒力基因和6种耐药基因.本研究分离得到1株肺炎克雷伯菌,通过血清型、药敏试验、全基因组分析表明,该菌株为高毒力且多药耐药菌株,与肺炎克雷伯菌的高致病性有关,具有感染人类和动物的潜在风险.
Klebsiella pneumoniae can cause pneumonia in mink.It has seriously affected the development of mink breeding industry.The purpose of this study was to analyze the virulence and drug resistance of Klebsiella pneumoniae from mink,and to provide thearetical basis for investigating the pathogenicity and drug resistance of Klebsiella pneumoniae.In this study,the bacteria were isolated and identified by pure culture,gram staining and PCR method.The agar microdilution method was used to detected the drug resistance spectrum of the strain.The serotype was identified by PCR method.The whole genome DNA was extracted and sequenced.The composition and important com-ponents of the whole genome sequence were analyzed.The genotypes,drug resistance genes and virulence genes of the strain were analyzed by bioinformatics of whole genome.The results showed that the isolated strain was Klebsiella pneumoniae,which belonged to K2 serotype.The results of drug sensitivity test suggested that the strain was resistant to tetracycline,chloramphenicol,florfenicol,ampicillin,ce-ftazidime,cefoxitin,ciprofloxacin,enrofloxacin and kanamycin,showing multiple drug resistance.The assembled genome size was 5435.522 kb and contained one chromosome and one plasmid.The whole genome analysis showed that the strain belonged to ST86.The strain carried 7 virulence genes and 6 resistance genes.In this study,one Klebsiella pneumoniae strain was isolated.Serotype,drug sensi-tivity test and whole genome analysis showed that the strain was highly virulent and multi-drug resistant,which was related to the high pa-thogenicity of Klebsiella pneumoniae and was easy to cause infection in humans and animals.
