蜜环菌是天麻生长发育的共生真菌,不同蜜环菌菌株对天麻产量及质量的影响差异较大,但由于其形态相似,限制了共生真菌菌株的快速选取.从知识产权保护的角度建立蜜环菌优良菌株的鉴定方法,具有必要性.本研究利用多位点序列分型(Multilocus se-quence typing,MLST)方法,基于市场上广泛应用的高卢蜜环菌的全基因组序列,设计了 10对特异性引物,对包括M3菌株在内的16株市售蜜环菌进行PCR检测.结合蜜环菌菌索发育特征构建菌株的鉴定方法,从16株蜜环菌中有效识别M3菌株.结果表明,利用10对特异性引物对16株蜜环菌进行PCR检测,发现12株蜜环菌的PCR检测结果各异,可以将它们与M3菌株区分开,剩3株蜜环菌菌株与M3菌株PCR结果相同.结合菌索发育形态发现,M3菌株菌索分支多且长,且远多于其余3株蜜环菌,因此,可以作为鉴定出M3菌株的特征.通过以上方法蜜环菌M3菌株可以从16个菌株中得以识别区分.基于多位点序列分型及菌索发育特点对蜜环菌进行比较鉴定,是一种可行的方法,对天麻共生优良蜜环菌的选择与保护具有重要意义.

Armillaria mellea is a symbiotic fungus crucial for the growth and development of Gastrodia elata.Variations in strains of Ar-millaria mellea significantly impact the yield and quality of G.elata.However,their morphological similarity hinders the rapid selection of symbiotic fungal strains.Therefore,it is imperative to establish an identification method for superior strains of Armillaria mellea from the perspective of intellectual property protection.This study employed the Multilocus Sequence Typing(MLST)method based on the whole-genome sequence of a high-yield Armillaria mellea in the market.Based on this sequence,we designed 10 pairs of specific primers for PCR detection of 16 commercially available Armillaria strains,including the M3 strain.A method for identifying strains was developed based on the developmental characteristics of Armillaria mellea rhizomorphs,effectively identifying the M3 strain among 16 strains of Armillaria mellea.The results indicated that,using 10 pairs of specific primers for PCR detection of 16 strains of Armillaria mellea,distinct PCR de-tection results were obtained for 12 strains,allowing them to be distinguished from the M3 strain.The remaining 3 strains of Armillaria mel-lea exhibited the same PCR results as the M3 strain.Based on the morphological development of fungal hyphae,it was observed that the M3 strain exhibits multiple and elongated fungal hyphae branches,significantly more than the other three strains of Armillaria.Hence,this can serve as a distinctive feature for identifying the M3 strain.Through the aforementioned methods,the M3 strain of Armillaria can be identified and differentiated from the other 16 strains.Comparative identification of Armillaria based on multi-locus sequence typing and fungal hyphae development characteristics is a viable method,which holds great significance for selecting and protecting excellent Armillaria strains sym-biotic with G.elata.
