铁死亡抵抗分子FSP1介导的食管癌5-氟尿嘧啶耐药机制及iFSP1化疗增敏作用的探究OA
Mechanism of FSP1-Mediated Ferroptosis Resistance in 5-Fluorouracil Chemoresistance of Esophageal Carcinoma and the Chemosensitization Effect of iFSP1
目的 探究铁死亡抑制蛋白 1(FSP1)在食管癌中的表达、与预后的关系及其介导 5-氟尿嘧啶(5-FU)化疗耐药的作用机制及 FSP1 抑制剂(iFSP1)的化疗增敏作用.方法 利用癌症基因组图谱(TCGA)分析FSP1 在食管癌中的表达及其与预后的关系;采用免疫组化检测 FSP1 在食管癌及癌旁组织中的表达;采用浓度梯度法培养 5-FU 耐药食管癌细胞;采用 Western blot 法检测耐药细胞 FSP1 蛋白的表达;采用细胞免疫荧光实验、流式细胞术、CCK-8 法、克隆形成实验检测 iFSP1 对食管癌化疗耐药的逆转作用.结果 FSP1 在食管癌组织中高表达且与不良预后相关;耐药食管癌细胞中 FSP1 蛋白表达量增高;FSP1 高表达与 5-FU 化疗耐药密切相关;使用FSP1 特异性抑制剂可提高肿瘤细胞铁死亡水平,并增强食管癌细胞对 5-FU 的敏感性.结论 FSP1 通过抑制肿瘤细胞铁死亡介导食管癌对 5-FU 的化疗耐药,靶向 FSP1 可能逆转食管癌的化疗耐药结局.
Objective To investigate the expression of ferroptosis suppressor protein 1(FSP1)in esophageal cancer,its relationship with prognosis,the mechanism by which it mediates 5-fluorouracil(5-FU)chemotherapy resistance,and the chemosensitizing effect of the FSP1 inhibitor(iFSP1).Methods The Cancer Genome Atlas(TCGA)was used to analyze FSP1 expression and its prognostic significance in esophageal cancer.Immunohistochemistry was performed to detect FSP1 expression in esophageal cancer and adjacent non-cancerous tissues.A 5-FU resistant esophageal cancer cell line was established using the concentration gradient method.Western blot was used to detect FSP1 protein expression in the resistant cells.Cell immunofluorescence,flow cytometry,CCK-8 assay,and colony formation assay were employed to evaluate the reversal effect of iFSP1 on chemoresistance in esophageal cancer.Results FSP1 was highly expressed in esophageal cancer tissues and was associated with poor prognosis.FSP1 protein expression was increased in 5-FU resistant esophageal cancer cells.High FSP1 expression was closely correlated with resistance to 5-FU chemotherapy.Treatment with a specific FSP1 inhibitor enhanced ferroptosis in tumor cells and increased the sensitivity of esophageal cancer cells to 5-FU.Conclusion FSP1 mediates 5-FU chemotherapy resistance in esophageal cancer by inhibiting ferroptosis in tumor cells.Targeting FSP1 may reverse chemotherapy resistance in esophageal cancer.
