目的 探讨 Pg 感染对食管鳞状细胞癌(ESCC)肺转移的影响,以及中性粒细胞胞外陷阱(NETs)在其中的作用.方法 将荧光素酶标记的 KYSE150 细胞(LKC)经尾静脉注射建立裸鼠 ESCC 肺转移模型.直接感染组(Pg 组)接种预先与Pg 共同培养的LKC,口腔暴露组(OR 组)在接种后用菌液涂抹口腔感染Pg,对照组(CON组)不感染.接种后第 3 周,每组进一步分为 GSK484(+)和 GSK484(-)两个亚组,前者腹腔注射 NETs 阻断剂GSK484,后者注射不含药物的溶剂.于第3 周(干预前)及第4~8 周每周行活体荧光成像,第8 周末取肺行离体荧光成像.收集 Pg 感染的中性粒细胞培养上清,以 Transwell 小室检测其对 KYSE30 细胞侵袭能力的影响.免疫组化检测 Ki67、Ly6G 表达;免疫荧光检测 MPO、Cit-H3 表达并行 DAPI 复染评价 NETs 形成;流式细胞术检测CD11b+、Ly6G+中性粒细胞比例.结果 Pg 组于第 3 周即出现肺部荧光信号,此后强度逐渐增强且最为显著,OR 组次之,CON 组无明显信号.Pg 感染中性粒细胞后的培养上清可显著增强KYSE30 细胞侵袭能力.Pg 组与OR 组肺转移灶内及周围 Ly6G+中性粒细胞显著富集,肺组织中性粒细胞比例升高,MPO 与 Cit-H3 表达增强且共定位增多.GSK484 干预可逆转上述变化,干预后肺组织中的中性粒细胞比例与 CON 组无显著差异.结论 Pg感染可促进ESCC 肺转移,该作用依赖肺组织中中性粒细胞的募集及NETs 形成;靶向NETs 形成可能是阻断ESCC肺转移的潜在策略.

Objective To investigate the effect of Porphyromonas gingivalis(Pg)infection on lung metastasis of esophageal squamous cell carcinoma(ESCC)and the role of neutrophil extracellular traps(NETs)during this metastatic process.Methods A nude mouse model of ESCC lung metastasis was established by tail vein injection of luciferase-labeled KYSE150 cells(LKC).In the direct infection group(Pg group),LKC pre-co-cultured with Pg were inoculated;in the oral exposure group(OR group),Pg bacterial suspension was applied to the oral cavity after inoculation;the control group(CON group)was not infected.At the 3rd week after inoculation,each group was further divided into two subgroups,GSK484(+)and GSK484(-),the former was intraperitoneally injected with the NETs inhibitor GSK484,and the latter was injected with drug-free solvent.In vivo fluorescence imaging was performed at the 3rd week(before intervention)and weekly from the 4th to the 8th week(experimental termination);ex vivo fluorescence imaging of the lungs was performed at the end of the 8th week.The culture supernatant of Pg-infected neutrophils was collected,and its effect on the invasive ability of KYSE30 cells was assessed using Transwell chambers.Immunohistochemistry was performed to detect Ki67 and Ly6G expression;immunofluorescence was used to detect myeloperoxidase(MPO)and citrullinated histone H3(Cit-H3)expression,and DAPI counterstaining was performed to evaluate NETs formation;flow cytometry was used to detect the proportion of CD11b+and Ly6G+neutrophils.Results Pulmonary fluorescence signals appeared in the Pg group as early as the 3rd week,gradually increased in intensity,and were the most prominent thereafter,followed by the OR group,whereas the CON group showed no obvious signals.The culture supernatant of neutrophils infected with Pg significantly enhanced the invasive ability of KYSE30 cells.Ly6G+neutrophils were significantly enriched within and around lung metastatic lesions in both the Pg and OR groups,the proportion of neutrophils in lung tissue was elevated,and MPO and Cit-H3 expression was enhanced with increased co-localization.GSK484 intervention reversed the above changes,and the proportion of neutrophils in lung tissue after intervention showed no significant difference compared with the CON group.Conclusion Pg infection promotes ESCC lung metastasis that depends on neutrophil recruitment and NETs formation in lung tissue,suggesting that targeting NETs formation may be a potential strategy to block ESCC lung metastasis.
