新菌株Devosia sp.JM3中DON脱氢酶的基因组挖掘和酶学性质表征OA
Genomic mining and enzymatic characterization of DON dehydrogenase from novel strain Devosia sp.JM3
试验旨在从新菌株德沃斯氏菌属(Devosia sp.)JM3的基因组中挖掘呕吐毒素(DON)脱毒关键脱氢酶,并表征其酶学性质.首先,运用生物信息学软件对Devosia sp.JM3新菌株进行基因组组装和基因预测,并通过序列比对完成对DON脱毒关键脱氢酶的基因组挖掘.其次,通过分子进化树构建、十二烷基硫酸钠聚丙烯酰胺凝胶电泳(SDS-PAGE)和高效液相色谱(HPLC)对其进行分析和DON脱毒效果验证.最后,探究温度、pH值、金属离子和DON初始浓度4个因素对该酶活性的影响.结果显示,挖掘到的目标蛋白与DON脱氢酶A(DepA)完全保守,其在64 kDa具有较高的表达量,在24 h内可将100 mg/L DON降解至原浓度的1.9%.在35℃、pH值7.5、底物初始浓度100 mg/L、1 mmol/L钙离子(Ca2+)参与的条件下,该酶对DON的转化率最高.研究表明,从Devosia sp.JM3菌株基因组中挖掘的DON脱氢酶是DepA,其酶学性质受温度、pH值、金属离子和底物初始浓度的影响.
The experiment aimed to mine the key detoxifying dehydrogenase for deoxynivalenol(DON)from the genome of the novel strain Devosia sp.JM3 and characterize its enzymatic properties.Firstly,bioinformatics software was used to perform genome assembly and gene prediction of the novel strain Devosia sp.JM3,and genome mining of the key DON detoxification dehydrogenase was completed through sequence alignment.Secondly,the enzyme was analyzed and validated for DON detoxification efficiency through molecular phylogenetic tree construction,sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE),and high-performance liquid chromatography(HPLC)detection.Finally,the effects of four factors,namely temperature,pH value,metal ions,and initial DON concentration,on the enzyme activity were investigated.The results showed that the target protein mined was completely conserved with the DON dehydrogenase A(DepA),exhibited high expression at 64 kDa,and could degrade 100 mg/L DON to 1.9%of its original concentration within 24 hours.Under the conditions of 35℃,pH value of 7.5,initial substrate concentration of 100 mg/L,and the presence of 1 mmol/L calcium ions(Ca2+),the enzyme achieved the highest conversion rate of DON.The study shows that the DON dehydrogenase mined from the genome of Devosia sp.JM3 is DepA,and its enzymatic properties are influenced by temperature,pH value,metal ions,and initial substrate concentration.
