石河子地区奶牛隐性乳房炎病原微生物的培养鉴定及微生物群落分析OA
Cultivation,identification,and microbial community analysis of pathogenic microorganisms causing subclinical mastitis in dairy cows in Shihezi area
试验旨在探究石河子地区奶牛隐性乳房炎(SCM)对牛乳微生物的影响.依据奶牛生产性能测定(DHI)数据筛选105份乳样,进行病原菌分离培养与PCR鉴定.根据鉴定结果分为金黄色葡萄球菌检出组(SG组)、链球菌检出组(GBS组)和对照组(HG组),每组6头,通过16S rRNA V3-V4区高通量测序分析3组乳样中微生物群落结构.结果显示,105份样品总体病原菌检出率为89.39%,金黄色葡萄球菌检出率(71.43%)高于其他病原菌.Alpha多样性分析显示,3组菌群物种丰富度与多样性差异不显著(P>0.05).线性判别分析效应大小(LEfSe)分析发现,GBS组中变形菌门富集,SG组与其他组无显著差异(P>0.05).功能预测发现,GBS组在嘌呤代谢、DNA修复与重组蛋白等通路显著富集,可能与病原菌定植和免疫逃避机制有关.研究表明,石河子地区奶牛患SCM的主要病原菌为金黄色葡萄球菌;两种病原菌引起的SCM均破坏了乳腺微生物环境,可能导致感染加剧.
The experiment aimed to investigate the impact of subclinical mastitis(SCM)in dairy cows in Shihezi area on milk microbiota.A total of 105 milk samples were screened based on the measurement of cow production performance(DHI)data,and pathogenic bacteria were isolated,cultured,and identified by PCR.According to the identification results,the samples were divided into three groups,namely Staphylococcus aureus detection group(group SG),Streptococcus detection group(group GBS),and control group(group HG),with six cows per group.The microbial community structure in the three groups of milk samples was analyzed by high-throughput sequencing of 16S rRNA V3-V4 regions.The results showed that the overall pathogen detection rate of 105 samples was 89.39%,and the detection rate of Staphylococcus aureus(71.43%)was higher than other pathogens.Alpha diversity analysis showed that there was no significant difference in species richness and diversity among the three groups(P>0.05).Linear discriminant analysis effect size(LEfSe)analysis revealed enrichment of Proteobacteria in the group GBS.There was no significant difference between SG group and other groups(P>0.05).Functional prediction revealed that the group GBS was significantly enriched in pathways such as purine metabolism,DNA repair and recombinant proteins,which may be related to pathogen colonization and immune evasion mechanisms.The study shows that the main pathogen causing SCM in dairy cows in Shihezi area is Staphylococcus aureus.Both types of pathogens causing SCM have disrupted the mammary microbiota environment,which may lead to exacerbation of infection.
