酸藤子活性物质蒽贝素对雌性大鼠中枢性性早熟调节作用OA
Regulating effect of embelin from Embelia laeta(L.)Mez on central precocious puberty in female rats
目的 探讨酸藤子活性物质蒽贝素对雌性大鼠中枢性性早熟(CPP)的调节作用.方法 从酸藤子干果粉中提取蒽贝素结晶,采用高效液相色谱面积比较法分析酸藤子提取蒽贝素纯度.36只雌性SD仔鼠随机分为正常对照组、性早熟模型组、亮丙瑞林组、蒽贝素高、中、低剂量组(EH、EM、EL),除正常组外,其余各组仔鼠采用达那唑诱导形成CPP模型,亮丙瑞林组和不同剂量蒽贝素组分别给予相应药物治疗.记录仔鼠体质量增长情况,ELISA法检测血清卵泡刺激素(FSH)和黄体刺激素(LH)水平,苏木精-尹红(HE)染色观察子宫形态学改变,免疫组织化学染色法观察下丘脑促性腺激素释放激素(GnRH)蛋白表达情况,Western blotting法检测垂体蛋白激酶C(PKC)和FSH-β蛋白表达情况.结果 亮丙瑞林、EH、EM组可抑制CPP仔鼠体质量增长,EL组直到30日龄时才可抑制CPP仔鼠体质量增长(P<0.05或P<0.01).与性早熟模型组相比,亮丙瑞林、EH、EM组仔鼠的血清FSH和LH水平降低,EL组仅可抑制CPP仔鼠血清FSH水平(P<0.05或P<0.01).与性早熟模型组相比,亮丙瑞林、EH、EM组仔鼠卵巢体积降低;亮丙瑞林组、蒽贝素各剂量组可下调CPP仔鼠下丘脑弓状核GnRH以及垂体PKC、FSH-β蛋白表达.结论 蒽贝素可以抑制CPP仔鼠体征改变,通过下调下丘脑弓状核GnRH以及垂体组织中PKC、FSH-β蛋白表达,从而调节血清LH、FSH水平,恢复性早熟仔鼠子宫、卵巢形态及功能.
Objective To investigate the regulatory effect of embelin on central precocious puberty in female rats,and to provide experimental basis for the development and application of anthracene in the corresponding fields.Methods The crystal of embelin was extracted from the dried fruit powder of Embelia laeta(L.)Mez.The purity of embelin extracted from Embelia laeta(L.)Mez was analyzed by HPLC with area comparison method.Thirty-six female SD rats(5-days-old)were randomly divided into normal control group,central precocious puberty model group,leuprorelin group with high,medium and low dose groups of embelin.Except for the normal group,the rats in the other groups were induced by danazol to form CPP model in 5-day-old female rats.The rats in the leuprorelin group and different doses of embelin group were treated with corresponding drugs.The weight gain of rats in each administration group was recorded.The regulation of serum FSH and LH were detected by ELISA.The morphological changes of uterus were observed by pathological HE staining.The expression of GnRH protein in hypothalamus was observed by immunohistochemical staining.The expression of PKC and FSH-β protein in pituitary tissues of rats was detected by Western blotting.Results Leuprorelin,high and medium doses of embelin could inhibit the weight gain of precocious puberty rats(P<0.05 or P<0.01).Low dose of embelin could inhibit the weight gain of precocious puberty rats until 30 days of age(P<0.05).The levels of serum FSH and LH in the leuprolide group,the high-dose and medium-dose embelin groups were significantly lower than those in the precocious puberty model group(P<0.05 or P<0.01).Low-dose of embelin could only inhibit the serum FSH level of precocious puberty offspring rats(P<0.05).Compared with the precocious puberty model group,the ovarian volume of the mice in the leuprorelin,high and medium dose groups was significantly reduced.The expression of GnRH protein in the hypothalamic arcuate nucleus of female rats in the leuprorelin group and the embelin intervention group was significantly down-regulated,which was significantly different from the precocious puberty model group.The expression of GnRH protein in the hypothalamic arcuate nucleus and PKC and FSH-β protein in pituitary tissues of precocious puberty rats were down-regulated by embelin.Conclusion Embelin can inhibit the changes of signs in central precocious puberty rats,regulate serum LH and FSH levels by down-regulating the expression of GnRH protein in hypothalamic arcuate nucleus,and restore the morphology and function of uterus and ovary in precocious puberty rats.
