使君子化学成分的提取分离及其抗氧化活性OA
Extraction and isolation of chemical constituents from Quispualis indica and their antioxidant activities
目的 提取分离使君子中的单体化合物并进行结构鉴定,然后探究其抗氧化作用.方法 将使君子药材粉碎,乙醇回流提取,合并滤液,将滤液分别用纯化水、正丁醇、乙酸乙酯、石油醚进行萃取,通过高效液相色谱C18柱分离出单体化合物,经纯度检测后利用理化性质与波谱解析鉴定出化合物的化学结构.然后以BHT为对照品,试验单体化合物清除DPPH·、ABTS·、O2-·、·OH这4种自由基的能力、与金属离子(Fe2+)的螯合能力、还原Fe3+能力,再研究它们对DNA间接损伤的保护作用.结果 从使君子药材中提取出4种单体化合物,经鉴定分别为白桦脂酸、没食子酸乙酯、盐酸葫芦巴碱与没食子酸.其中没食子酸、没食子酸乙酯有清除DPPH·、ABTS·、O2-·、·OH这4种自由基的能力,还有与金属离子(Fe2+)螯合能力及还原Fe3+能力上表现出了较好的抗氧化能力;白桦脂酸有清除ABTS·、O2-·、·OH这3种自由基的能力,还有与金属离子(Fe2+)螯合能力及还原Fe3+能力上表现出了较好的抗氧化能力;而盐酸葫芦巴碱与金属离子(Fe2+)螯合能力最强,其抗氧化能力相对另外3种单体化合物表现较弱.4种单体化合物抗氧化能力均与化合物浓度呈正相关关系,且对羟基自由基间接损伤DNA都有一定的保护作用.结论 从使君子中提取出的4种单体化合物都具有一定的抗氧化能力,其中没食子酸和没食子酸乙酯的抗氧化能力表现较好,白桦脂酸和盐酸葫芦巴碱的抗氧化能力表现较弱,且4种单体化合物抗氧化能力与化合物浓度有一定正相关性.
Objective The monomeric compounds in Quispualis indica were extracted and their structures were identified,and then their antioxidant effects were explored.Methods Pulverize Quisqualis indica,extract with ethanol under reflux,combine the filtrates,extract the filtrates with purified water,n-butanol,ethyl acetate and petroleum ether respectively,and separate the monomer compounds by high performance liquid chromatography C18 column.After detection,the chemical structure of the compound was identified by physicochemical properties and spectral analysis.Then,with BHT as the reference substance,the ability of the monomer compounds to scavenge DPPH·,ABTS·,O2-·,·OH,the chelating ability with metal ions(Fe2+),and the ability to reduce Fe3+were tested,and their protective effect on indirect DNA damage was studied.Results Four kinds of monomeric compounds were extracted from the Quispualis Indica,which were identified as betulinic acid,ethyl gallate,trigonelline hydrochloride and gallic acid.Among them,gallic acid and ethyl gallate scavenging DPPH·,ABTS·,O2-·,·OH these four free radicals,as well as with the metal ion(Fe2+)chelating ability and reducing Fe3+ability to show a good antioxidant capacity.Betulinic acid showed good antioxidant capacity in scavenging ABTS·,O2-·,·OH free radicals,chelating ability with metal ions(Fe2+)and reducing Fe3+ability.The chelation ability of trigonelline hydrochloride with metal ions(Fe2+)was the strongest,and its antioxidant capacity was weaker than that of the other three monomer compounds.The antioxidant capacity of the four monomer compounds was positively correlated with the concentration of the compound,and had a certain protective effect on the indirect damage of DNA by hydroxyl radicals.Conclusion The four monomeric compounds extracted from quispualis indica have a certain antioxidant capacity,among which the antioxidant capacity of gallic acid and ethyl gallate is better,the antioxidant capacity of betulinic acid and trigonelline hydrochloride is weaker,and the antioxidant capacity of the four monomeric compounds has a certain positive correlation with the compound concentration.
