宁夏枸杞内生菌分离和鉴定OA
Isolation and Identification of Endophytic Microorganisms in Lycium barbarum L.
枸杞(Lycium barbarum L.)是我国重要的药食同源植物,富含内生菌资源.以'宁杞1号'组培苗为研究对象,通过内生菌分离、培养纯化、菌落形态学观察、DNA提取、PCR扩增、测序及NCBI数据库比对等方法,系统地分离鉴定其内生菌群落,并比较不同长势植株的内生菌群落差异.经分离鉴定共获得7种内生菌,菌群组成特征显示,链格孢属(相对丰度38.2%)为优势真菌,鞘氨醇单胞菌属(相对丰度19.1%)为优势细菌;并发现鞘氨醇单胞菌与甲基杆菌的丰度与宿主长势密切相关,其在"优势单株"中的检出率(11.7%)显著高于"劣势单株"(4.3%).研究结果表明,'宁杞1号'组培苗中含有丰富的内生菌资源,且其群落结构,尤其是有益菌群的丰度,与宿主的生长活力密切相关,这为深入研究内生菌与枸杞的共生关系、开发新型微生物菌剂奠定了理论基础.
Lycium barbarum L.is an important medicinal and edible plant in China,rich in endophytic microbial resources.Taking the tissue culture seedlings of'Ningqi No.1'as the research object,through methods such as endophytic microbial isolation,cultivation purification,colony morphological observation,DNA extraction,PCR amplification,sequencing and comparison with the NCBI database,the endophytic microbial community of these seedlings was systematically isolated and identified.The differences in the endophytic microbial communities among different growth stages of the plants were also compared.Through isolation and identification,a total of 7 endophytic microorganisms were obtained.The composition characteristics of the microbial community showed that Alternaria Nees(relative abundance 38.2%)was the dominant fungus,and Sphingomonas(relative abundance 19.1%)was the dominant bacterium.It was also found that the abundance of Sphingomonas and Methylobacterium was closely related to the growth vigor of the host,and the detection rate of these microorganisms in the"dominant individual"(11.7%)was significantly higher than that in the"inferior individual"(4.3%).The research results indicated that the tissue culture seedlings of'Ningqi No.1'contained abundant endophytic microbial resources,and their community structure,especially the abundance of beneficial microorganisms,was closely related to the growth vitality of the host.This lays a theoretical foundation for in-depth research on the symbiotic relationship between endophytic microorganisms and Lycium barbarum and the development of new microbial agents.
