韭菜白腐病病原菌的分离与鉴定OA
Isolation and Identification of the Pathogen Causing White Rot Disease of Chinese Chive
为明确甘肃省定西市临洮县大棚韭菜菌核病的病原菌种类,为当地病害防控提供病原学依据,本研究采集当地典型发病韭菜病样,分离纯化获得菌株25-4-GSJC,采用离体叶片接种、活体盆栽土壤接种结合人工回接验证病原菌致病性,并通过形态学观察、ITS序列PCR扩增与测序及系统发育分析等方法鉴定病原菌.结果显示,该菌株经划伤接种与盆栽菌土接种均可使韭菜表现出田间典型发病症状;分离回接菌株与原始菌株菌落形态特征一致,满足柯赫氏证病法则;其在PDA培养基上菌丝呈白色,不产生分生孢子,可形成黑色致密小型菌核;通过PCR扩增获得长度814 bp的ITS目的基因片段,序列同源比对及系统发育树分析显示,供试菌株与白腐小核菌(Stromatinia cepivora)模式菌株聚类于同一进化分支,自举支持率达96%.综上,本研究首次确证了该地区大棚韭菜菌核病的病原菌为白腐小核菌(Stromatinia cepivora).
To identify the pathogen species causing sclerotinia disease in greenhouse chives in Lintao county,Dingxi city,Gansu province,and to provide an etiological basis for local disease control,this study collected typical diseased Chinese chive samples from the local area.A strain designated 25-4-GSJC was isolated and purified.Pathogenicity was confirmed through in vitro leaf inoculation,in vivo potted soil inoculation,and artificial re-inoculation.The pathogen was identified using morphological observation,ITS sequence PCR amplification and sequencing,and phylogenetic analysis.The results showed that both wound inoculation and potted soil inoculation induced typical field disease symptoms on Chinese chives.The re-isolated strain from inoculated plants exhibited the same colony morphology as the original strain,satisfying Koch's postulates.On PDA medium,the mycelium was white,no conidia were produced,and small,dense black sclerotia were formed.An 814 bp ITS gene fragment was amplified by PCR.Sequence homology and phylogenetic tree analysis indicated that the tested strain clustered with the type strain of Stromatinia cepivora in the same evolutionary clade with 96%bootstrap support.In conclusion,the pathogen causing sclerotinia disease in greenhouse Chinese chives in this region is firstly identified as Stromatinia cepivora.
