弥漫性中线胶质瘤中PP1α和p-ERK的表达及其与临床病理特征和预后的关系OA
The expression of PP1α and p-ERK in diffuse midline glioma tissues and its relationship with clinicopathological characteristics and prognosis
目的 探讨弥漫性中线胶质瘤(diffuse midline gliomas,DMG)伴H3K27变异型组织中PP1α、p-ERK及肿瘤侵袭相关因子MMP2、MMP9蛋白表达与临床病理特征及预后的关系.方法 收集31例DMG和9例正常脑组织石蜡包埋样本,采用免疫组化法检测PP1α、p-ERK、MMP2、MMP9的表达及其与临床病理特征的关系.结果 31例DMG组织中PP1α、p-ERK、MMP2、MMP9的表达高于对照组.PP1α高表达与年龄≤18岁(P=0.044)、ATRX蛋白表达缺失(P=0.020)相关;p-ERK高表达与年龄≤18岁(P=0.018)相关;PP1α高表达与p-ERK表达(C=0.380,P=0.035)呈中等强度关联,且p-ERK高表达与MMP2表达呈中等强度关联(C=0.470,P=0.003);Kaplan-Meier曲线显示PP1α高表达(P=0.002)、p-ERK高表达(P=0.028)、患者年龄≤18岁(P=0.039)、肿瘤发生于非丘脑中线部位(P=0.027)、组织学表现为高级别(P=0.019)、术后未行辅助治疗(P=0.002)及p53表达≥10%(P<0.01)患者预后较差;Cox多因素分析提示:PP1α高表达(HR:4.661;95%CI:1.219~17.825;P=0.024)、术后未行辅助治疗(HR:1.768;95%CI:0.456~6.857;P=0.041)及p53表达≥10%(HR:4.233;95%CI:1.528~11.722;P=0.011)是影响DMG患者预后的独立危险因素.结论 DMG组织中PP1α和p-ERK呈高表达,且两者表达具有相关性,PP1α高表达患者预后较差,PP1α可能在DMG肿瘤恶性进展中发挥重要作用.
Objective To investigate the relationship between the expression levels of PP1α,p-ERK and tumor invasion-related factors MMP2 and MMP9 in diffuse midline gliomas(DMG)with H3K27 alteration tissue and clinico-pathological features and prognosis.Methods 31 cases of DMG and 9 cases of normal brain tissue paraffin-embedded samples was collected.Immunohistochemistry was uesd to detect the expression of PP1α,p-ERK,MMP2,and MMP9.Relationship between PP1α,p-ERK,MMP2,and MMP9 expression and clinical pathological characteristics was analyzed.Results The expression levels of PP1α,p-ERK,MMP2 and MMP9 in 31 cases of DMG tissues were higher than those in the control group.High expression of PP1α was associated with age≤18 years(P=0.044)and loss expression of ATRX protein(P=0.020);high expression of p-ERK was associated with age≤18 years(P=0.018);high expression of PP1α was moderately correlated with the expression of p-ERK(C=0.380,P=0.035),and high expression of p-ERK was moderately correlated with the expression of MMP2(C=0.470,P=0.003).The Kaplan-Meier curve showed that patients with high expression of PP1α(P=0.002),high expression of p-ERK(P=0.028),age≤18 years(P=0.039),tumor located in non-thalamic midline region(P=0.027),histological grade of high level(P=0.019),no postoperative adjuvant therapy(P=0.002),and p53 expression≥10%(P<0.01)had a poorer prognosis.Cox multivariate analysis suggested that high expression of PP1α(HR:4.661;95%CI:1.219-17.825;P=0.024),no postoperative adjuvant therapy(HR:1.768;95%CI:0.456-6.857;P=0.041),and p53 expression≥10%(HR:4.233,95%CI:1.528-11.722;P=0.011)were independent risk factors affecting the prog-nosis of DMG patients.Conclusion PP1α and p-ERK is highly expressed in DMG tissues,and their expressions are correlated.Patients with high expression of PP1α have poor prognosis,and PP1α may play an important role in the ma-lignant progression of DMG tumors.
