为揭示宁夏贺兰山东麓不同小产区赤霞珠葡萄的代谢差异及"风土"调控机制,选取该产区6个代表性酒庄的10年生赤霞珠为研究对象,以美贺酒庄(M1)为对照,采用液相色谱-质谱联用(LC-MS)非靶向代谢组学技术,分析各酒庄与美贺酒庄葡萄浆果的代谢物组成差异,筛选关键差异代谢物并开展KEGG通路注释分析.结果表明,共检测到9类3 546个代谢物,其中有机氧化合物占比最高(11.48%),脂肪酰类(10.77%)、羧酸及其衍生物(9.93%)和类黄酮(9.31%)次之;6个酒庄与美贺酒庄葡萄浆果的显著性差异代谢物集中在羧酸及其衍生物、类黄酮和有机氧化合物类;KEGG通路富集分析显示,仁益源酒庄(M2)、立兰酒庄(M3)与美贺酒庄(M1)差异最显著的通路为氧化磷酸化,玉泉营酒庄(M4)与美贺酒庄(M1)为氨基酰-tRNA生物合成,维嘉妮酒庄(M5)与美贺酒庄(M1)为柠檬酸循环(TCA循环),西鸽酒庄(M6)、罗兰马歌酒庄(M7)与美贺酒庄(M1)为果糖和甘露糖代谢.研究明确了宁夏贺兰山东麓不同产区赤霞珠葡萄浆果的核心差异代谢物类别及关键代谢通路,为解析该产区酿酒葡萄"风土"调控机制、实现品质精准调控提供理论依据.

To reveal the metabolic differences of Cabernet Sauvignon grapes from different sub-regions of the Helan Mountain's Eastern Foothill Wine Region of Ningxia and the regulatory mechanism of"terroir",ten-year-old Cabernet Sauvignon grapes from six representative wineries were selected as the research objects.The Meihe winery(M1)in the region was used as the control.Using liquid chromatography-mass spectrometry(LC-MS)non-targeted metabolomics technology,the differences in the composition of metabolites between the grape berries of these wineries and those of the Meihe winery were analyzed.Key different metabolites were screened and KEGG pathway annotation analysis was conducted.The results showed that a total of 9 categories of 3,546 metabolites were detected,among which organic oxidized compounds accounted for the highest proportion(11.48%),followed by fatty acids(10.77%),carboxylic acids and their derivatives(9.93%),and flavonoids(9.31%);The significant difference metabolites of the grape berries of the six wineries and the Meihe winery were mainly carboxylic acids and their derivatives,flavonoids,and organic oxidized compounds;KEGG pathway enrichment analysis showed that the most significantly different pathways between Renyiyuan winery(M2),Lilan winery(M3),and Meihe winery(M1)were oxidative phosphorylation,those between Yuquanying winery(M4)and Meihe winery(M1)were aminoacyl-tRNA biosynthesis,between Weijiani winery(M5)and Meihe winery(M1)were citric acid cycle(TCA cycle),and between Xige winery(M6),Luolanmage winery(M7)and Meihe winery(M1)were fructose and mannose metabolism.The study clarified the core different metabolite categories and key metabolic pathways of Cabernet Sauvignon grape berries from different sub-regions of the Helan Mountain's Eastern Foothill Wine Region of Ningxia,providing a theoretical basis for analyzing the"terroir"regulation mechanism of the wine grapes in this region and achieving precise quality regulation.
