博落回内生真菌Penicillium turbatum的次级代谢产物OA
Secondary Metabolites of Endophytic Fungus Penicillium turbatum from Macleaya cordata(Willd.)R.Br.
为了研究博落回内生真菌Penicillium turbatum的次级代谢产物,采用马铃薯葡萄糖液体培养基发酵培养,综合运用 HP20大孔树脂、MCI GEL CHP20P大孔树脂层析和半制备 HPLC等色谱手段对代谢产物进行分离纯化,并在此基础上探究代谢产物对5种细菌的抑菌活性.结果表明,共分离得到11个单体化合物,分别是青霉酸、二氢青霉酸、苔黑酚、苔色酸、吲哚甲醛、吲哚-3-乙酸甲酯、4,4'-二羟基二苯甲酮、4-(2-羟乙基)苯酚、3-甲氧基-4-甲基-3,5二氢-1H-吡咯、鸟嘌呤核苷、腺苷,均为首次从Penicillium turbatum次级代谢产物中分离得到;青霉酸对紫色杆菌、大肠埃希氏杆菌、枯草芽孢杆菌和金黄色葡萄球菌均表现出抑菌活性,最小抑菌浓度分别为25,50,100,100 mg/L;二氢青霉酸对金黄色葡萄球菌有一定抑制作用,最小抑菌浓度为100 mg/L.
In order to research the secondary metabolitesof the endophytic fungus Penicillium turbatum from Bolehui,the fermentation was carried out using a potato glucose liquid medium.Comprehensive chromatographic methods such as HP20 macroporous resin,MCI GEL CHP20P macroporous resin chro-matography,and semi-preparative HPLC were employed to separate and purify the metabolites.On the basis of that,the antibacterial activities of the metabolites against five types of bacteria were further in-vestigated.The results showed that a total of 11 monomeric compounds were isolated,namely penicillic acid,dihydropenicillic acid,orcinol,orsellinic acid,1H-Indole-3-carbaldehyde,Methyl Indole-3-acetate,4,4'-dihydroxybenzophenone,4-(2-hydroxyethyl)phenol,3-methoxy-4-methyl-3,5-dihydro-1H-pyrrol-2-one,guanosine,and adenosine.The 11 compounds were obtained from Penicillium turbatum for the first time.Penicillic acid has certain inhibitory effect on Chromobacterium violaceum,Escherichia coli,Bacil-lus subtilis and Staphylococcus aureus,minimum inhibitory concentration values were 25,50,100 and 100 mg/L respectively.Dihydropenicillic acid has certain inhibitory effect on Staphylococcus aureus,and its minimum inhibitory concentration value is 100 mg/L.
