小分子药物在疾病治疗中发挥着重要作用,但对于很多小分子抗癌药物来说,普遍存在着靶向性差、溶解度低和稳定性差等缺点,这些都严重限制了药物的临床应用.小分子前药自组装纳米制剂在克服药物的这些缺点方面发挥着重要作用.通过总结近年来小分子前药自组装纳米制剂最新进展,自组装结构理论基础、基于前药结构的分类、相对于传统纳米颗粒的优势、"智能"刺激响应释放系统、多领域联用以及未来挑战等多个方面,旨在为该领域的科研人员提供有价值的参考与启示,推动小分子自组装前药技术的持续创新与发展.

Small molecule drugs play an important role in the treatment of diseases.For many small molecule anticancer drugs,there are widespread shortcomings such as poor targeting,low solubility and poor stability,which seriously limit the clinical application of drugs.Small molecule prodrug self-assembly nanopreparations play an important role in overcoming these shortcomings of drugs.This paper summarizes the latest progress of small-molecule prodrug self-assembly nanopreparations in recent years,including the theoretical basis of self-assembly structure,classification based on prodrug structure,advantages over traditional nanoparticles,"intelligent"stimulus-response release system,multi-domain application and future challenges,etc.,aiming to provide valuable reference and enlightenment for researchers in this field.Promote the continuous innovation and development of small molecule self-assembly prodrug technology.
