基于肠道菌群探究补中益气汤治疗肌肉减少症的作用及机制OA
Effect and mechanism of Buzhong Yiqi Decoction in the treatment of sarcopenia based on gut microbiota
目的 探讨补中益气汤对肌肉减少症的治疗作用及其对肠道菌群的调控机制.方法 将32只7月龄的雄性C57BL/6小鼠随机分为空白组、模型组、补中益气汤组和营养支持组,每组8只.空白组给予等体积生理盐水,其余3组均皮下注射地塞米松(DXM,5 mg/kg,0.1 mL/10 g)以建立肌肉减少症模型.给药过程中,空白组和模型组均给予等体积生理盐水灌胃,补中益气汤组给予补中益气汤8.19 g/(kg·d)灌胃,营养支持组给予亮氨酸0.520 g/(kg·d)及骨化三醇0.065 μg/(kg·d)灌胃,连续8周.观察小鼠行为及运动功能,采用超高频小动物超声检测腓肠肌横截面积,应用16S rRNA测序分析肠道菌群.结果 与空白组相比,模型组小鼠疲劳转棒耐受时间及腓肠肌横截面积分别减少10.49 s和0.68 mm2(P<0.05);与模型组相比,补中益气汤组上述指标分别增加8.48 s和0.59 mm2(P<0.05);营养支持组上述指标分别增加8.45 s和0.56 mm2(P<0.05),改善程度略小于补中益气汤组.肠道菌群分析显示,与模型相比,补中益气汤组厚壁菌门与拟杆菌门的比值升高,乳杆菌属与毛螺梭菌属丰度降低,杜氏杆菌属丰度升高,阿克曼菌属显著富集;营养支持组仅部分调节上述菌群.结论 补中益气汤对肌肉减少症模型小鼠具有显著的治疗效果,能增加腓肠肌肌肉含量,延长疲劳耐受时间,改善运动能力,其补中益气、健脾强肌的作用机制可能与调节肠道菌群相关,为从肠道菌群角度阐释补中益气汤改善肌肉减少症的作用提供了实验依据.
Objective To investigate the therapeutic effect of Buzhong Yiqi Decoction on sar-copenia and its regulatory mechanism on gut microbiota.Methods Thirty-two 7-month-old male C57BL/6 mice were randomly divided into blank group,model group,Buzhong Yiqi Decoction group,and nutritional support group,with eight mice in each group.The blank group received an e-qual volume of normal saline,while the other three groups were subcutaneously injected with dexam-ethasone(DXM,5 mg/kg,0.1 mL/10 g)to establish a sarcopenia model.During the treatment pe-riod,the blank and model groups received an equal volume of normal saline by gavage;the Buzhong Yiqi Decoction group received Buzhong Yiqi Decoction at 8.19 g/(kg·d)by gavage;and the nutri-tional support group received leucine at 0.520 g/(kg·d)and calcitriol at 0.065 μg/(kg·d)by gavage,for eight consecutive weeks.Behavioral and motor functions were observed.Gastrocnemius muscle cross-sectional area was measured using ultra-high-frequency small-animal ultrasound,and gut microbiota composition was analyzed by 16S rRNA sequencing.Results Compared with the blank group,the model group showed a reduction of 10.49 s in the fatigue rotarod endurance time and a decrease of 0.68 mm2 in gastrocnemius muscle cross-sectional area(P<0.05).Compared with the model group,the Buzhong Yiqi Decoction group showed increases of 8.48 s and 0.59 mm2 in the aforementioned indices,respectively(P<0.05);the nutritional support group showed increases of 8.45 s and 0.56 mm2,respectively(P<0.05),although the degree of improvement was slightly less than that in the Buzhong Yiqi Decoction group.Gutmicrobiota analysis revealed that,compared with the model group,the Buzhong Yiqi Decoction group exhibited an increased ratio of Firmicutes to Bacteroidetes,reduced abundances of Lactobacillus and Lachnoclostridium,increased abundance of Dubosiella,and significant enrichment of Akkermansia;the nutritional support group only partially modulated the aforementioned microbiota.Conclusion Buzhong Yiqi Decoction exerts a significant therapeutic effect on sarcopenia model mice by increasing gastrocnemius muscle mass,prolonging fa-tigue endurance time,and improving motor function.The underlying mechanism of replenishing qi and strengthening the spleen and muscle may be associated with the modulation of gut microbiota.This study provides experimental evidence for elucidating the role of Buzhong Yiqi Decoction in a-meliorating sarcopenia from the perspective of gut microbiota.
