醉鱼草发酵前后总黄酮含量及其抗菌活性OA
Content of Total Flavonoids and Their Antibacterial Activity in Buddleja lindleyana Fort.Before and After Fermentation
目 的 探讨发酵对醉鱼草总黄酮含量及其抗菌活性的影响.方 法 采用加热回流提取法对醉鱼草中的总黄酮进行提取;采用紫外分光光度法,以芦丁为对照品,以 5%NaNO2、5%Al(NO3)3、4%NaOH为显色剂,在510 nm波长处,对醉鱼草发酵前后总黄酮含量进行测定;以枯草芽孢杆菌对醉鱼草总黄酮进行发酵,采用单因素试验法优化醉鱼草总黄酮的发酵时间、发酵pH以及接菌量;采用金黄色葡萄球菌和大肠杆菌为供试菌,以抑菌圈大小为指标对发酵前后抗菌活性变化进行比较.结 果 发酵前总黄酮含量为(0.114 2±0.004 0)g/g,发酵后总黄酮含量为(0.102 0±0.002 2)g/g;发酵后总黄酮抗菌活性大于发酵前.利用 SPSS 22.0软件对结果进行单因素 ANOVA统计分析,结果表明发酵前后总黄酮的含量存在显著性差异;发酵前后总黄酮对大肠杆菌抑菌活性存在极其显著性差异,而对金黄色葡萄球菌抑菌活性存在显著性差异.结 论 研究表明醉鱼草发酵前后总黄酮含量有明显变化,同时发酵后比发酵前总黄酮的抑菌活性增强.为中药材发酵后药用价值的研究提供参考价值,也可为中药材发酵物的客观评价提供参考.
Objective To explore the effect of fermentation on the total flavonoid content of Buddleja lindleyana Fort.(B.lindleyana)and its antibacterial activity.Methods Total flavonoids from B.lindleyana were extracted by the heat reflux extraction method.The total flavonoid contents before and after fermentation were determined by UV spectrophotometry at 510 nm,using rutin as the reference substance and 5%NaNO2,5%Al(NO3)3,and 4%NaOH as color-developing agents.Bacillus subtilis was used to ferment the total flavonoids of B.lindleyana,and the fermentation time,fermentation pH,and inoculum size were optimized by single-factor experiments.Staphylococcus aureus and Escherichia coli were used as test strains,and changes in antibacterial activity before and after fermentation were compared using the diameter of the inhibition zone as the indicator.Results The total flavonoids content before fermentation was(0.114 2±0.004 0)g/g,while that after fermentation was(0.102 0±0.002 2)g/g;The antibacterial activities of total flavonoids after fermentation was greater than that before fermentation.One-way ANOVA was performed using SPSS 22.0,and the results showed that there was a significant difference in total flavonoid content before and after fermentation.There was a highly significant difference in the antibacterial activity of total flavonoids against Escherichia coli before and after fermentation,and a significant difference in their antibacterial activity against Staphylococcus aureus.Conclusion This study showed that the total flavonoid content of B.lindleyana changed significantly before and after fermentation,and that the antibacterial activity of total flavonoids was enhanced after fermentation.This study provides a reference for research on the medicinal value of fermented traditional Chinese medicinal materials and also for their objective evaluation.
