通阳活血方对慢性心力衰竭大鼠心功能和血清炎症因子TNF-α、IL-6的影响OA
Effects of Tongyang Huoxue Formula on cardiac function and serum inflammatory factors TNF-α and IL-6 in rats with chronic heart failure
目的 观察通阳活血方对慢性心力衰竭(CHF)大鼠心功能和血清炎症因子肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)的影响,探讨通阳活血方治疗CHF可能的作用机制.方法 52只SD大鼠随机分为假手术组(10 只)、手术组(42 只),手术组采用腹主动脉缩窄法制备CHF模型,造模成功后随机分为模型组、通阳活血方组、比索洛尔组,各 14只.通阳活血方组灌胃通阳活血方颗粒溶液(21.25 g·kg-1·d-1);比索洛尔组灌胃富马酸比索洛尔混悬液(1 mg·kg-1·d-1);假手术组和模型组灌胃等量蒸馏水.造模后 1 周开始灌胃给药,每日 1 次,至造模后 8 周.造模后 16 周实验周期结束,检测超声心动图、血清氨基末端脑利钠肽前体(NT-proBNP)水平、TNF-α水平、IL-6 水平及心脏质量指数,HE染色、Masson染色观察心肌组织病理变化.结果 与假手术组比较,模型组大鼠收缩末期内径(LVESD)、舒张末期内径(LVEDD)增加(P<0.05),左室射血分数(LVEF)、左心室短轴缩短率(LVFS)降低(P<0.05),NT-proBNP、TNF-α、IL-6 水平升高(P<0.05,P<0.01),心脏质量指数升高(P<0.05).与模型组比较,通阳活血方组与比索洛尔组大鼠LVESD、LVEDD均减少(P<0.05),LVEF均升高(P<0.05),NT-proBNP水平降低(P<0.05,P<0.01),心脏质量指数降低(P<0.05);通阳活血方组TNF-α、IL-6 水平降低(P<0.05,P<0.01);比索洛尔组IL-6 水平降低(P<0.01).结论 通阳活血方可减轻CHF大鼠心肌肥厚,改善心肌纤维化,降低血清NT-proBNP水平,改善心功能,抑制血清炎症因子TNF-α、IL-6的过度表达.
Objective To observe the effects of Tongyang Huoxue Formula(TYHXF)on cardiac function and serum inflamm-atory factors TNF-α and IL-6 in rats with chronic heart failure(CHF),and to explore the possible mechanism of action of TYHXF in treating CHF.Methods Fifty-two SD rats were randomly divided into sham operation group(n=10),and operation group(n=42).The CHF model was established in the surgical group by abdominal aortic constriction.After successful modeling,rats were randomly assigned into model,TYHXF,and bisoprolol groups,with 14 rats in each group.The TYHXF group received TYHXF granule(21.25 g·kg-1·d-1)by gavage;the bisoprolol group received bisoprolol fumarate oral suspension(1 mg·kg-1·d-1)by gavage;and both the sham operation and model groups received an equal volume of distilled water by gavage.Gavage administration started one week after modeling,and continued once daily until eight consecutive weeks.At the end of the 16-week experimental period after modeling,echocardiography was performed,levels of serum N-terminal pro-B-type natriuretic peptide,TNF-α and IL-6,and cardiac mass index were measured.Histopathological changes in myocardial tissue were observed by HE and Masson staining.Results Compared with the sham operation group,rats in the model group exhibited increased left ventricular end-systolic diameter(LVESD)and left ventricular end-diastolic diameter(LVEDD)(P<0.05),decreased left ventricular ejection fraction(LVEF)and left ventricular fractional shortening(LVFS)(P<0.05),elevated levels of NT-proBNP,TNF-α and IL-6(P<0.05,P<0.01),and increased cardiac mass index(P<0.05).Compared with the model group,both the TYHXF and bisoprolol groups showed reduced LVESD and LVEDD(P<0.05),increased LVEF(P<0.05),decreased NT-proBNP levels(P<0.05,P<0.01),and decreased cardiac mass index(P<0.05).The TYHXF group showed reduced levels of TNF-α and IL-6(P<0.05,P<0.01),and the bisoprolol group showed reduced IL-6 levels(P<0.01).Conclusion TYHXF can alleviate myocardial hypertrophy,improve myocardial fibrosis,reduce serum NT-proBNP levels,improve cardiac function,and inhibit the overexpression of serum inflammatory factors TNF-α and IL-6 in rats with CHF.
