妊娠前半期甲状腺功能减退症的代谢特征及生物标志物OA
Metabolic characteristics and biomarkers of hypothyroidism in the first half of pregnancy
目的:探究妊娠前半期甲状腺功能减退症(简称甲减)的代谢特征与生物标志物.方法:选择妊娠前半期甲减孕妇30 例(甲减组)和正常孕妇30 例(对照组).收集孕妇血清样本,采用高分辨液相质谱技术进行非靶向代谢组学检测.采用正交偏最小二乘判别分析筛选甲减组与对照组的差异代谢物.对差异代谢物进行 KEGG 通路富集分析.结果:甲减组与对照组共有323 个差异代谢物,与对照组相比,甲减组中上调的差异代谢物 96 个,下调的差异代谢物226 个.与对照组比较,甲减组 13-羟基十八碳二烯酸(13-HODE)下调,鞘氨醇(Sph)上调(P<0.05).差异代谢物主要富集于PPAR 信号通路、细胞凋亡等10 条通路.结论:妊娠前半期甲减血清代谢特征发生改变,13-HODE 与 Sph 可能是潜在的生物标记物.
Aim:To explore the metabolic characteristics and potential biomarkers of hypothyroidism in the first half of pregnancy.Methods:A total of 30 pregnant women with hypothyroidism in the first half of pregnancy were selected as the hypothyroidism group,and another 30 healthy pregnant women were enrolled as the control group.Serum samples of all preg-nant women were collected,and untargeted metabolomics detection was performed by high-resolution liquid chromatography-mass spectrometry.Orthogonal partial least squares-discriminant analysis(OPLS-DA)was adopted to screen differential me-tabolites between the hypothyroidism group and the control group.KEGG pathway enrichment analysis was carried out for the screened differential metabolites.Results:A total of 323 differential metabolites were identified between the hypothyroidism group and the control group.Compared with the control group,96 differential metabolites were up-regulated and 226 were down-regulated in the hypothyroidism group.Compared with the control group,13-hydroxyoctadecadienoic acid(13-HODE)was significantly down-regulated,while sphingosine(Sph)was significantly up-regulated in the hypothyroidism group(P<0.05).The differential metabolites were mainly enriched in 10 pathways,such as the PPAR signaling pathway and apoptosis pathway.Conclusion:The serum metabolic characteristics are altered in patients with hypothyroidism during the first half of pregnancy.13-HODE and Sph may serve as potential biomarkers for this condition.
