Optimization of Process Conditions for Enzyme-Assisted-Ultrasonic Synergistic Extraction of Sugarcane Polyphenols

以甘蔗皮为原料,采用酶辅助-超声协同提取技术(EASE)提取多酚类物质.在单因素实验基础上,运用Box-Behnken响应面法优化纤维素酶添加量、超声功率、提取温度和超声时间4个关键工艺参数.方差分析表明,各因素对多酚得率的影响程度依次为:超声功率>纤维素酶添加量>提取温度>超声时间,建立的二次多项式回归模型拟合度良好(R2=0.9746).最优工艺条件为:纤维素酶添加量0.45%、超声功率350 W、提取温度52 ℃、超声时间35 min,在此条件下多酚得率为4.79 mg/g±0.05 mg/g.与传统热回流提取法和单一超声辅助提取法相比,协同提取时间明显缩短,多酚得率分别提高28.4%和12.4%,DPPH自由基清除率也有所提升.酶解破壁与超声空化传质具有显著互补效应,可有效促进甘蔗皮多酚的释放.本研究为甘蔗加工副产物的高值化利用及功能性食品原料开发提供了技术支撑.

Using sugarcane rind as raw material,polyphenols were extracted by enzyme-assisted ultrasonic synergistic extraction(EASE).On the basis of single-factor experiments,the Box-Behnken response surface methodology was employed to optimize four key process parameters:cellulase dosage,ultrasonic power,extraction temperature,and ultrasonic time.Analysis of variance showed that the influence of the factors on polyphenol yield followed the order of ultrasonic power>cellulase dosage>extraction temperature>ultrasonic time,and the established quadratic polynomial regression model exhibited a good fit(R2=0.9746).The optimal process conditions were cellulase dosage of 0.45%,ultrasonic power of 350 W,extraction temperature of 52 ℃,and ultrasonic time of 35 min.Under these conditions,the polyphenol yield reached 4.79 mg/g±0.05 mg/g.Compared with traditional heat reflux extraction and single ultrasound-assisted extraction,the synergistic extraction significantly shortened the extraction time,and increased the polyphenol yield by 28.5%and 12.3%,respectively;the DPPH radical scavenging activity was also improved.The complementary effects between enzymatic cell wall disruption and ultrasonic cavitation-enhanced mass transfer effectively promoted the release of sugarcane rind polyphenols.This study provides technical support for the high-value utilization of sugarcane processing by-products and the development of functional food ingredients.
