鱼腥草对急性肺损伤大鼠肺组织的保护作用OA
Protective effects of Houttuynia cordata Thunb.on lung tissue of rats with acute lung injury
目的:探讨鱼腥草(HcT)对急性肺损伤(ALI)大鼠肺组织的保护作用及可能机制.方法:将 SD 大鼠随机分为6 组,正常对照组、模型组、地塞米松组(5 mg/kg),HcT 低剂量组(7.2 g/kg)、中剂量组(14.4 g/kg)、高剂量组(28.8 g/kg),每组10 只,连续灌胃给药5 d(1 次/d)后,除正常对照组外,其余5 组均予腹腔注射脂多糖5 mg/kg诱导 ALI.脂多糖注射6h 后记录大鼠生理状况,测定肺系数,HE 染色观察肺组织病理学改变,ELISA 法检测血清、肺泡灌洗液和肺组织中 TNF-α、IL-1β、IL-6 和 IL-10 的浓度,检测肺组织中谷胱甘肽过氧化物酶(GSH-Px)活性及总抗氧化能力(T-AOC);RT-qPCR 法检测肺组织中 NLRP3、TNFAIP3、NFKBIA 和 NF-κBp65 mRNA 的表达,West-ern blot 法检测 NLRP3、NF-κB 和 NF-κBp65 蛋白的表达.结果:与模型组比较,各 HcT 组大鼠肺系数降低,肺组织炎症细胞浸润减少,肺泡隔增厚和肺泡水肿情况均有所改善,ALI 病理评分下降,血清、肺泡灌洗液和肺组织中 IL-1β、IL-6、TNF-α 浓度,肺组织中 NLRP3、TNFAIP3、NFKBIA、NF-κBp65 mRNA 表达,NLRP3、NF-κB 和 NF-κBp65 蛋白表达降低,GSH-Px 活性、T-AOC 和 IL-10 浓度升高(P 均<0.05);且高剂量组变化最显著(P 均<0.05).结论:HcT可能通过调控 NLRP3/NF-κB 通路,增强抗氧化应激能力和抑制内质网应激,抑制炎症反应,从而减轻脂多糖诱导的大鼠 ALI.
Aim:To investigate the protective effects of Houttuynia cordata Thunb.(HcT)on lung tissue of rats with a-cute lung injury(ALI)and to explore the underlying mechanisms.Methods:Sixty SD rats were randomly divided into 6 groups:normal control group,model group,dexamethasone group(5 mg/kg),low-dose HcT group(7.2 g/kg),medium-dose HcT group(14.4 g/kg),and high-dose HcT group(28.8 g/kg),with 10 rats in each group.Rats were administered the cor-responding treatment by gavage once daily for 5 consecutive days.Except the normal control group,rats in the other 5 groups received an intraperitoneal injection of lipopolysaccharide(LPS)5 mg/kg to induce ALI.Six hours after LPS injection,phys-iological status was recorded,and the lung index was measured.Histopathological changes in lung tissue were observed by HE staining.The levels of TNF-α,IL-1β,IL-6,and IL-10 in serum,bronchoalveolar lavage fluid(BALF),and lung tissue were determined by ELISA method.Glutathione peroxidase(GSH-Px)activity and total antioxidant capacity(T-AOC)in lung tissue were also measured.The mRNA expression of NLRP3,TNFAIP3,NFKBIA,and NF-κBp65 in lung tissue were detected by RT-qPCR,and the protein expression of NLRP3,NF-κB,and NF-κBp65 were analyzed by Western blot.Re-sults:Compared with the model group,the lung index and inflammatory cell infiltration reduced,alleviated alveolar septal thickening and pulmonary edema alleviated,and the pathological injury score decreased in 3 HcT groups.In addition,the levels of IL-1β,IL-6,and TNF-α in serum,BALF,and lung tissue decreased,and the mRNA expression of NLRP3,TN-FAIP3,NFKBIA,and NF-κBp65 and the protein expression of NLRP3,NF-κB,and NF-κBp65 downregulated;GSH-Px ac-tivity,T-AOC,and IL-10 levels increased(all P<0.05).These effects were most pronounced in the high-dose HcT group(all P<0.05).Conclusion:HcT may alleviate LPS-induced ALI in rats by enhancing antioxidant capacity and suppressing inflammatory responses,possibly through regulation of the NLRP3/NF-κB signaling pathway.
